Study On Pharmacokinetics Of Cefoperazone Sodium And Tazobactam Sodium For Injection

To estabilish an High performance liquid chromatography ( HPLC ) method to monitor concentrations of Cefoperazone (CPZ)and Tazobactam (TB) in human plasma and urine sample, to assess for pharmacokinetics after adminstration Cefoperazone Sodium and Tazobactam Sodium for Injection (6:1) (CPZ/TB) in healthy volunteers, and to provide scientific evidence for reasonable guideline for clinical applicaton.Results and Conclusion:1. A simple rapid and sencitive analytical method for the determination of CPZ and TB was developed. The separation of CPZ was performed on a Diamonsil C18( 150×4.6mm, 5μm) column and the mobile phase consisting of (containing 20mmol·L^-1 NaH₂PO₄, pH=3.0) water: methanol ( 65/35 ,v/v).The flow rate was 1mL·min^-1; the detection wavelength was set at 254nm. The separation of TB on a Diamonsil C18 column(150×4.6mm,5μm).The mobile phase consisting of (containing 20 mmol·mL^-1 NaH2PO4 and 0.5 mmol·L^-1 Tetrabutylammonium bromide, pH=6.5 ) water: acetonitrile (84/16, v/v).The flow rate was 1mL·min-1; the detection wavelength was set at 230nm. The good validation criteria results of the method and sensitivity allowed its use to determine CPZ and TB in human plasma and urine sample.2. Blood drug level increased with the dose of CPZ/TB, the pharmacokinetic parameters(α,β, t_1/2α, t_1/2β, Cls)of CPZ and TB with all doses were simalar. The mean plasma concentrations - time curve after a administration was coincident with a two–compartment open model. CPZ and TB had not pharmacokinetic interaction significantly.3. There was a significant correlation between the AUC of CPZ and TB and administration doses. There was a significant correlation between the C_max of CPZ and TB and administration doses. The pharmacokinetic character of CPZ and TB accord with linearity pharmacokinetics.
