| To improve the drug targeting and reduce the side effect of paclitaxel, the paclitaxel-HA freeze-drying preparation was prepared. As a drug carrier, HA can controll the sustained durg release, target to cancer cells, and increase the bioavailability of paclitaxel. In this paper, in vitro activity and lipidomics analysis of paclitaxel-hyaluronan preparation were studied.In vitro inhibition activity study of paclitaxel-HA preparation on Hela cell indicated that the inhibitory rate can reach about 80%. Preparation with different molecular weight of HA showed no significant difference of inhibitory rate. Considering the stability and dissolution study of preparation, the optimal result is HA-31w preparation. The HA-31w preparation incuced apoptosis of Hela cell directly with a arrest of G2/M phase of cell cycle, the apoptosis rate can up to 56.8% at 24h.The phospholipid profile of Hela cell has been established by LC/ESI/MSn approach, there are over 112 glycerophospholipid molecular species of 6 different glycerophospholipid classes in Hela cells. Under the effect of preparation over 6h, the quantitative analysis of glycerophospholipid showed significant increase in the abundance of PG, PE, PI and PS, and no significant differences in the abundance of PC and PA. |
