Objective The purpose of the present was to investigate the effect of FGF-4 on the proliferation and differentiaion of bone marrow-derived mesemesenchymal stem cells (MSCs) into ligaments cells and the effect of the scleraxis on it.Methods In the current study,the principles of tiuuse engineering were producing cultured MSCs with the bFGF,in which bFGF had multiple biological efficacies to accelerate the proliferation and differentiaion of MSCs.With the low dose FGF-4(200 ng/μl)in cultures, then the MSCs were measured by histochemistry ,MTT,RT-PCR.Results Besides promotion of proliferation of MSCs obviousely,and the synthesis of collagen 1,collagen3 ,tenascin ,which were essential extracellular matrix proteins and cytoskeletal elements of ligaments and tendons,were express highly after the activation of scleraxis .Conclusion MSCs with FGF-4 increases its proliferation and synthesis of essential extracellular matrix proteins and differentiates into tenocytes.This new tissue engineering approach could be of potential to enhance the repaire of damaged ligaments or tendons, especially those caused by diseases during the advanced stages.
