Study On Antitumor Action Of "Xiaoliutang" Recipe

Objective:"XiaoLiuTang"recipe is clinically an effective prescription to cure many patients with tumour, but the mechanism has not been interpreted. In this studies, we choose the human HepG2 cells, employing zoopery and serum- pharmacology to discuss the mechanism of"XiaoLiuTang"recipe.Methods: To make a mode of implanted tumour with injecting HepG2 cells in the oxter of mice.①The effect of inhibitory in humour tissue with the method of irragating stamach of the white rat with"XiaoLiuTang"recipe was observed.②The effect of inhi- bitory in HepG2 by"XiaoLiuTang"recipe with the method of MTT was investigated.③The apoptosis of HepG2 was measured by Flow cytometry- (FCM).④The expression of caspase-3 and bcl-2 was investigated by immunohistochemistrty.⑤To see the Telomerase activity of HepG2 cells with the method of PCR-ELISA.RESULTS:①The bulk of groups of medication was smaller than the normal saline group's, especially the big-dosage group's(the inhibitatory rate is 41.88%).②The results of the MTT show that the inhibitory rate of HepG2 cells and the medication dosage were in linear relation. On the third day the rate reaches its max that is 52.24%.③the FCM indicated that the big-dosage"XiaoLiuTang"recipe serum group had significant accelerating effect after three days and the rate of apoptosis was 22.3%,and the difference between the big-dosage group and the control group has statistical significance④ the immunohistochemistrty results show that expression of the caspase-3 was increased and bcl-2 was decreased in a concentration-dependent manner.⑤Fourteen days later the big-dosage medication group have significant inhibitory effect on telomerase activity of hepatocarcinoma.Conclusion:"XiaoLiuTang"recipe can inhibit the hepatocarcinoma, also can induce humour cells to apoptosis. The mechanism may be that"XiaoLiuTang"recipe restores some tumor suppressor genes expression and increases the expression of caspase-3 while decreased expression of bcl-2,also may correlate to inhibiting telomerase activity.
