Part I Study of the distribution of Nav1.1 in guinea-pig ventricular cellsObjective To observe Nav1.1 in the isolation of guinea-pig ventricular cells with laser scanning confocal microscope.Methods Ventricular cells were isolated from adult guinea-pig in the Langendorff preparation. The isolated adult guinea-pig ventricular cells were divided into three groups: control group, antibody group, antibody and triton group. The guinea-pig ventricular cells underwent labeling of Nav1.1 by immunofluorescence technique after isolation, then were observed with laser scanning confocal microscope.Results Nav1.1 was observed in the cell membrane without triton and in the cytoplasm of adult guinea-pig ventricular cells with triton.Conclusions Laser scanning confocal microscopic observation reveals that Nav1.1 is expressed in the guinea-pig ventricular cells. Part II Effect of anti-Nav1.1 antibody on sodium current in adult guinea-pig ventricular cellsObjective To examine the effect of anti-Nav1.1 antibody on sodium current in adult guinea-pig ventricular cells.Methods Ventricular cells were isolated from adult guinea-pig in the Langendorff preparation. The isolated adult guinea-pig ventricular cells were divided into four groups: control group, 1:100 antibody group, 1:50 antibody group, 1:25 antibody group. With whole cell patch clamp recording, we detected the sodium current in ventricular cells after applying anti-Nav1.1 antibody to the isolated guinea-pig ventricular cells.Results Anti-Nav1.1 decreased the voltage activated Na+ currents in a concentration dependent manner. The density of Na+ current of control was [(34.22±6.22) pA/pF, n=8], while those of test were ([(32.13±3.45) pA/pF, n=9], [(25.49±7.4) pA/pF, n=8] and [(19.23±1.69) pA/pF, n=9] by anti-Nav1.1 at 1:100; 1:50; 1:25, respectively. Sodium current is inhibited by 1:25 anti-Nav1.1 significantly. 1:25 anti-Nav1.1 shifted I-V relationship curve to more positive potentials.Conclusions Anti-Nav1.1 antibody can inhibit Na+ currents.
