Yupingfeng(YPF),which has been collected in the Pharmacopoeia of People's Republic of China,is the classic prescription of "reinforcing qi and consolidating the exterior" and is also one kind of the basic Chinese patent medicine.YPF is composed by Radix Astragali(RA),Rhizoma Atractylodes Macrocephale(RAM) and Radix Saposhnikoviae(RS).By means of documents statistics we find that the composition proportions of YPF are as many as twelve kinds,and the classic proportion is 3:1:1,which is recorded in Chinese Pharmacopoeia,and is used to treat people who suffers from exterior deficiency and weakness,spontaneous perspiration due to heat wind or susceptibility of pathogenic wind.Another representative proportion of 1:1:1,from the "Golden Mirror of Medicine",is used for "pathogenic wind stasis and continuing bleeding";proportion of 2:2:1,from "Yi Fang Lei Ju", is for the "people whose interspaces of skin and muscles are loose and easy flu";proportion of 4:4:1,from the "Yi Xue Cong Zhong Lu",is for spontaneous perspiration belonging to Yang deficiency and YPF should be used with the addition of oysters and floating wheat in order to Biao Shi Bu Yang.In this paper,the contents of the effective components in YPF with different compatibility proportion(dcp) are studied,based on the theory of prescriptions and pharmacology.The main research work of our project includes the following aspects:firstly,a method of water extraction(WE) and herb residue ethanol extraction(EE) is established,and then the content of the main effective components are determined by UV spectrophotometry, HPLC,AAS et al.The results show that in the four intact recipes and the three separated recipes the content of total polysaccharide>total amino acid>total flavonoids>total saponins.The content of total polysaccharide is relatively higher in the group of RA and RAM,and is the highest in RAM. Total amino acid,total saponins,astragalosideⅣ,quercetin and formononetin are mainly from RA and the contents of quercetin and formononetin are increasing with the increase of RA. 5-o-methylvisammiosode and prim-o-glucasylcimifugin are the active components in RS and the content of the former is greater than the latter in RS.They are up to the highest amount in the proportion of 1:1:1 due to their maximum contents in RS and are down to the lowest in the proportion of 4:4:1.The results of determination of trace elements indicate that in RA the content of iron is the highest and in RS the other four elements are the highest.In addition,the solubility in water of total polysaccharide,total flavonoids,total saponins,quertein,5-o-methylvisammiosode and prim-o-glucasylcimifugin are greater than their solubility in ethanol,whereas the total amino acid,astragalosideⅣand formononetin are on the contrary.In the four intact recipes,the ordering of the sum of total polysaccharide, total fiavonoids,total saponins total amino acid,and the sum of total effective components without trace elements,and the sum of total effective components are all 2:2:1>1:1:1>3:1:1>4:4:1,but the ordering of the sum of four kinds of flavonoids is 1:1:1>3:1:1>2:2:1>4:4:1.The contents of total polysaccharide,total saponins,5-o-methylvisammiosode, prim-o-glucasylcimifugin,copper and magnesium are the highest in the proportion of 1:1:1,while quercetin,formononeitin and the sum of five trace elements are the highest in the proportin of 3:1:1.
