[Objective]To explore the role of transfected p21waflgene in the duplication of centrosomes and the proliferation of human breast carcinoma cells.[Methods]p21waflgene packaged with lipofectin was transfected into the cells of a human breast carcinoma cell line MCF-7,which stably expressed p21wafl gene The cell growth was detected by using MTT.The cell cycle pattern was assayed by using flowcytometry.The duplication of centrosomes of breast carcinoma cells were detected by using indirect immunofluorescence microscopy.[Results]After transfection of pIRES-p21wafl,The p21wafl mRNA and protein of MCF-7-p21waflwere highly expressed as compared with MCF-7 cells.Cell growth was obviously inhibited and resulted in an accumulation of cells in G1,presenting that the proportion of the cells in G1 phase was obviously increased from(49.84±1.58)%to (70.52±3.21)%,and that of the cells in S and G2/M phases was obviously decreased from (36.83±1.36)%to(16.38±1.17)%and from(15.64±1.09)%to(9.42±0.98)%,resp -ectively, as compared with control group.The proportion of cells which contained unnormal centrosomes in MCF-7-p21waflwere obviously decreased as compared with MCF-7 cells (P<0.01).[Conclusion]The transfer of p21wafl gene could inhibit the growth and the unnormal duplication of centrosomes of human breast carcinoma cells,which even could inhibit the genesis and development of breast cancer and might be become a new method to treat the breast cancer.
