Objective To explore the procedures of isolation,cultivation and purification of endothelial progenitor cells(EPC) from peripheral bloods of rats,and to provide a new cell source for the treatment of ischemic myocardial injury using stell cell transplantation.Methods The Sprague-Dawley(SD) rats were injected rG-CSF subcutaneous for 7 days,then the peripheral bloods were obtained,mononuclear cells of peripheral bloods were isolated by Ficoll density gradient centrifugation,then the cells were cultured and amplified in special culture solution which included in 20% low-glucose IMDM,fetal bovine serum,horse serum,vascular endothelial growth factor,and basic fibroblast growth factor.The culture cells were identified by immunofluorescein stain with CD31,CD34,F1k-1,and vWF,the abilities of binding UEA-1 and taking ac-LDL were detected.Results The culture cells were identified and confirmed to endothelial progenitor cells by immunofluorescein stain with CD31,CD34,F1k-1 and vWF,and these cells had the abilities of binding UEA-1 and taking Ac-LDL.Conclusion Endothelial progenitor cells could be obtained successfully by obtaining mononuclear cells from the peripheral bloods of the rG-CSF mobilized SD rats and culturing in vitro,and EPC could be a new cell source for the treatment of ischemic myocardial injury using stell cell transplantation.
