Objective:To study the toxicity,target site and histomorphological changes of Artesunate to Aedes albopictus's 4th larvae.Methods:1.To observe the toxicity of Artesunate to Aedes albopictus's 4th larvae.2.To determine the median lethal concentration(LC50) of Artesunate to Aedes albopictus's 4th larvae.3.To observe the histomorphological changes of Aedes albopictus's 4th larvae by HE staining.4.To observe the changes of body surface of Aedes albopictus's 4th larvae treated by Artesunate through SEM.5.To observe the genomic DNA changes of Aedes albopictus's 4th larvae by agarose gel electrophoresis.Results:1.Under Artesunate,Aedes albopictus's 4th larvae showed toxic symptom of the bouche biting anal gills,atrophy of body and abdominal darkening. 2.The higher the concentration of Artesunate were,the stronger the toxicity of Artesunate to Aedes albopictus's 4th larvae was;the longer Aedes albopictus's 4th larvae was treated by Artesunate,the higher the mortality of Aedes albopictus's 4th larvae was.3.The median lethal concentration(LC50) of Artesunate to Aedes albopictus's 4th larvae was 6.64mg/ml.4.The histological study of Aedes albopictus's 4th larvae treated by Artesunate showed that the midguts were shrinkage,some vacuoles can be observed in the midintestine cell,striated borders was destroyed and peritrophic membrane had disappeared.5.The observation with scanning electron microscope showed that the body surfaces of Aedes albopictus's 4th larvae treated by Artesunate were shrinkage,disrupted and damaged.6.The agarose gel electrophoresis results showed that genomic DNA of the dead Aedes albopictus's 4th larvae treated by Artesunate(9mg/ml) appear "ladder".Conclusion:1.The toxicity of Artesunate on Aedes albopictus's 4th larvae was evident.2.The median lethal concentration(LC50) of Artesunate to Aedes albopictus's 4th larvae was 6.64mg/ml.3.The results of histological study showed that the tissue targets of Artesunate were midgut and body surface;the toxicity of Artesunate on Aedes albopictus's 4th larvae was mainly through contacting action and stomach action.4.In a certain concentration,Artesunate can induce apoptosis of Aedes albopictus's 4th larvae.
