Crude polysaccharides from the fruit body of Pleurotus abalonus were isolated by series of sequential steps including extraction with hot water,precipitation with 95% ethanol,removal of protein by Sevag method.The water-soluble polysaccharide(PPA-2) were obtained using DEAE Sephadex A-25 anion-exchange chromatography and Sephadex G-100 gel filtration chromatography.PPA-2 was homogeneous identified by polyacrylamide gel electrophoresis with a molecular mass of 23000.The infrared spectrum analysis showed that PPA-2 had polysaccharides specific absorbent peak,and the specific absorbent peak of nucleic acid and protein were not be found in the UV spectrum.A certain dosage of PPA-2 could inhibit the transplanted tumor growth of S180, improve non-specific immune function and specific immune function such as humoral immunity,cellular immunity for mice.Vivo experiments showed that PPA-2(75 mg/kg,100 mg/kg) have antitumor activities on the implanted Sarcoma 180 of mice(P<0.01).PPA-2(50 mg/kg,75 mg/kg, 100 mg/kg) could increase the spleen index in tumor-bearing mice(P<0.01,P<0.001). PPA-2(75 mg/kg,100 mg/kg) could increase the spleen index in normal mice(P<0.05,P<0.01).PPA-2(50 mg/kg,75 mg/kg,100 mg/kg) could increase the spleen index in lower immune mice(P<0.05,P<0.01,P<0.001),and enhance the phagocytic rate(P<0.05,P<0.01) and the phagocytic index(P<0.05,P<0.001) of celiac macrophage, production of serum hemolysins IgM(P<0.05) in normal mice.PPA-2(75 mg/kg,100 mg/kg) could enhance the function of delay hypersensitivity induced by DNFB(P<0.05, P<0.001) to normal mice.
