BackgroundColorectal cancers, including colon and rectal cancer, are the most common malignant tumor of digestive tract. Its fatality rate in malignant tumors is the third,and its incidence has increased recent years. Due to its strong invasiveness and metastasis,colorectal cancer can lead to body damage, organ failure, and even death. As the fight against tumor invasion and metastasis , we find that extracellular matrix (ECM) and degradation of biological barrier damage have played a role at the tumor invasion and metastasis. Matrix metalloproteinase -9 (MMP-9) can degrade extracellular matrix and collagen type IV. Cancerous cells make a breakthrough happen at primary site, invasion and metastasis. CD-147, also known as extracellular matrix metalloproteinase inducer, has been wide concerned by a variety of ways to stimulate production and release of MMPs. The relevance between MMP-9 and CD-147 becomes hot spot areas in drug research of antagonistic tumor, which may inhibit tumor invasion and metastasis.Objective:To investigate the expression of CD-147, MMP-9 in colorectal cancer tissues and its relationship between tumor growth, invasion, and metastasis .At the same time , to explore its relationship between CD-147 and MMP-9 with clinicapathological parameters. To provide new ideas for early diagnosis,prognosis assessment and treatment in colorectal cancers.Methods:43 cases material were studied from the second hospital of JiLin University during March 2007 to Octorber 2008,which were confirmed colorectal cancers by pathology. We used immunohistochemical SP staining method, X2 test, SPSS13.0 statistical analysis software packages.Results:1.The expression of CD-147, MMP-9 in colorectal cancer, para-cancer tissues, normal tissues.The positive rates of CD-147 were 31.03% 44.83%, 82.76% in normal tissues, adjacent tissues and cancer tissues respectively. There were significant difference (P <0.01). The positive rates of MMP-9 were 24.13%, 43.10%, 79.31% in normal tissue, adjacent tissues and cancer tissues (P <0.01).2.The relationship between CD-147 , MMP-9 and age, gender.Positive rates of CD-147 were 82.86%, 82.61% in men and women (P> 0.05). Age≥60, age <60 years , the positive rates of CD-147 was 81.08%, 85.71% in patients with colorectal cancer. There was no significant difference(P> 0.05). The positive rates of MMP-9 in colorectal cancer between men and women were 80.00%, 78.26%. There were no significant difference (P> 0.05). The positive expression rates were 78.39%, 80.95% in Age≥60 and age <60 years. There were no significant difference (P> 0.05). We can conclude that there was no cottelation between wxpression of CD-147 , MMP-9 and gender age(p>0.05).3.The relationship between CD-147, MMP-9 expression in colorectal cancer and the clinicopathological parameters.The positive rates of CD-147 in high / medium, poorly differentiated type were 82.92%, 82.35%. There was no significant difference (P> 0.05). The positive expression rate was 11.11% for the shallow and 95.23% for deep, 96.43% for full-thickness. There were a significant difference between three tissues(P <0.01). The positive rate CD-147 with lymph node metastasis was 95.83%, which significantly higher than that without lymph node metastasis ( 73.53%) (P <0.05). The positive rates of MMP-9 in the high / medium, poorly differentiated type were 78.49%, 82.35% respectively. There was no significant difference (P> 0.05). The positive rates of different degrees of invasion were 11.11% for the shallow , 90.48 % for deep, 92.86% for full-thickness. There was significant difference between the expression of MMP-9 and tissue distribution (P <0.01). The positive rates of MMP-9 in carcinoma of lymph node metastasis was 95.83%, which significantly higher than that without lymph node metastasis ( 67.65%) (P <0.01).4.The relationship between the expression of CD-147 and MMP-9 in colorectal cancer.In 58 cases of colorectal cancer, the positive rate of CD-147 and MMP-9 was 72.41% (42/58). The negative rate was 27.59% (16/58). There was a trend that the expression of MMP-9 increases according to CD-147. There was positive correlation between CD-147 and MMP-9 (γ= 0.4325, P = 0.032 <0.05).Conclusion:1.The espressions of CD-147 were higher than adjacent tissue,normal tissue(p<0.05).2.The expressions of CD-147 and MMP-9 were related to tumor infiltration, lymph node metastasis.But there were no relationship between the expressions of CD-147,MMP-9 and age,sex, tumor's differentiation ,TNM staging. 3.There was positive correlation between the expression of CD-147 and MMP-9. We can conclude that CD147 way be a stimulating factor for MMP-9.4.CD-147 and MMP-9 may be the clinical monitoring indicators, such as the early diagnosis of colorectal cancer, measure the degree of malignancy, determine prognosis, and so on.
