Objectives:To observe the expression ofβ2-AR, M3-AChR and GR in the bronchial airway of the asthma mouse model, evaluate the effects of beta2-agonist, muscarinic receptor antagonist and ICS on airway remodeling in the asthma mouse model, and their effects on the expression ofβ2-AR, M3-AChR and GR respectively.Methods:All fifty female BALB/c mice were offered by animal experimental center of preclinical medical college of Jilin University. Mice were randomly divided into five groups (each consisting of ten mice): normal control group(Group DZ), model group(Group MX), terbutaline group(Group TB), ipratropium bromide group (Group YB) and budesonide group(Group BD). The chronic asthmatic model was established by the sensitization and challenged of ovalbumin at the concentration of 0.2% and 2.5% respectively. The level of airway remodeling was observed by means of hematoxylin and eosin staining. The expression ofβ2-AR, M3-AChR and GR in bronchial airway were detected by means of immunohistochemistry. The change of airway wall, smooth muscle and inner wall were observed by computer-assisted image analysis software.Result:1. The results of airway remodeling:⑴Ratio between the area of airway wall and circumference of basement membrane of Group DZ, MX, TB, YB and BD were (12.89±1.71), (25.37±4.25), (15.98±2.58), (15.82±2.91), (13.86±2.21).The thickness of total airway wall in Group MX were significantly greater than Group DZ, TB, YB and BD (P﹤0.01), and the thickness of total airway wall in Group TB, YB, BD and DZ had no difference (P﹥0.05).⑵Ratio between the area of smooth muscle and circumference of basement membrane of Group DZ, MX, TB, YB and BD were (2.55±0.72), (7.58±2.16), (3.54±1.63), (3.36±1.69), ( 3.00±1.18). The thickness of smooth muscle in Group MX were greater than Group DZ, TB, YB and BD (P﹤0.05), and the thickness of smooth muscle in Group TB, YB, BD and DZ had no difference (P﹥0.05). 2. The result of the expression of three kinds receptor:⑴Compared with the Group DZ, the expression ofβ2-AR in bronchial airway of Group MX and TB was significantly lower(P﹤0.01).⑵The expression of M3-AChR in bronchial airway of Group MX and DZ had no significantly difference (P﹥0.05). Compared with the Group DZ, the expression of M3-AChR in Group YB was higher (P﹤0.05).⑶Compared with the Group DZ, the expression of GR in bronchial airway of Group MX was significantly lower (P﹤0.01), and the expression of GR in bronchial airway of Group BD was lower(P﹤0.05). Conclusions:1. The asthma mouse model was successfully established by the sensitization and challenged of ovalbumin at the concentration of 0.2% and 2.5% respectively.2. Early inhaled beta2-agonist, muscarinic receptor antagonist and ICS could efficiently alleviate the level of airway remodeling of the asthma mouse model. The effect of three kinds of antasthmatic on the proliferation of airway smooth muscle had no difference.3. The expression ofβ2-AR was down-regulated in remodeled bronchial airway of asthma mouse model, and inhaled beta2-agonist for a long time could down-regulate the expression ofβ2-AR in bronchial airway of asthma mouse model.4. There was not a visible change of the M3-AChR's expression in remodeled bronchial airway of asthma mouse model, and inhaled muscarinic receptor antagonist for a long time could up-regulate the expression of M3-AChR in bronchial airway of asthma mouse model.5. The expression of GR was down-regulated in remodeled bronchial airway of asthma mouse model, and inhaled ICS for a long time could down-regulate the expression of GR in bronchial airway of asthma mouse model.
