The Baisc Effect Of Gene FLJ22349 | | Posted on:2009-11-27 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y Y Dai | Full Text:PDF | | GTID:2144360272482089 | Subject:Biochemistry and Molecular Biology | | Abstract/Summary: | PDF Full Text Request | | BACKGROUND & OBJECTIVE:Human hypothetical protein FLJ22349 is located on chromosome 22q13.2 and encodes 229 amino acids.Biological information analysis suggested wide expression profile of FLJ22349 mRNA in human cancer tissues.This study was to predict the biological information of human FLJ22349 by Bioinformatics software and investigate the basic effect on cell lines.METHODS: Prediction the function of FLJ22349 was examined through different biomedical databases.The expressions of the FLJ22349 mRNA in mouse tissues,human cancer cell lines and HEK293 were detected by reverse transcription polymerase chain reaction(RT-PCR);the recombinant eukaryotic expression vector containing wide-type FLJ22349 was transfected into HEK293,MCF-7,and MDA-MB231 cell lines by cation polymex.The stable transfected cells were selected by G418 and amplified.The effect of FLJ22349 on cell growth was assessed by MTT assay. Subcellular distribution of human FLJ22349 was detected under fluorescent microscope.RESULTS:FLJ22349 is conserved across diverse species,including human,chimpanzee,rhesus,rat,mouse,dog,and cow,and shares no obvious homology to any known genes or protein.RT-PCR analysis revealed FLJ22349 gene expression in many mouse tissues and human cancer cell lines.FLJ22349 is mainly localized in the nucleus.The cell growth rates in HEK293/pcDNA3.1(+)-FLJ22349 and MCF-7/ pcDNA3.1(+)-FLJ22349 cell lines were higher than HEK293/ pcDNA3.1(+) and MCF-7/ pcDNA3.1(+) respectively(P<0.05).After 120h FLJ22349 transfection,the growth rates of HEK293 and MCF-7 were 299%,456% while those controlled group were 233%,272%.CONCLUTION:FLJ22349 is a protein that increases cells growth when over expressed in HEK293 and MCF-7 cell lines. [Purpose]To evaluate the association between SNP309 in MDM2(Murine double minute 2) promoter and breast cancer susceptibility.[Methods]Searched on CBM disc and PUBMED to get the case-control study on the association between SNP309 and breast cancer susceptibility,and pooled the OR of the association of SNP309 and breast cancer susceptibility by meta-analysis.Then we performed familial breast cancer subgroup analysis and sensitivity analysis and publication bias test.[Results]10 studies with 7535 cases and 8272 controls were enrolled in this meta-analysis.The pooled OR of G allele compared to T allele is 1.06 without statistically significant association(95%CI:0.96-1.06).There were no differences in MDM2 SNP309 frequencies among familial breast cancer cases.[Conclusion]The 309 T>G polymorphism in MDM2 was not statistically significant association with the breast cancer susceptibility. | | Keywords/Search Tags: | Breast cancer cell lines, HEK293, FLJ22349 gene, Cell growth, MDM2, SNP309, Breast cancer, meta-analysis | PDF Full Text Request | Related items |
