The main characteristics of tumor cells is the growing out of control and the ability of invasion and metastasis, and these two both involved with the non-contact inhibitory growth behaviors of tumor cells .It has been shown that the tumor cells non-contact inhibitory growth had two kinds of forms, that is, within tumor tissue the tumor cells were disorderly piled up, tumor cells which in border area expanded and invaded to the normal tissue in vivo. In vitro culture for the tumor cell proliferation fast it was easy to grow into multi-storey overlap. The essential of the overlapping growth was the result of the imbalance growth and cell migration out of control. To analysis the formation of overlapping colony helped us to highlight the formation and mechanism of the un-contact inhibitory growth in tumor cells.Objective: To study the tumor cells groups obstacle to social behaviors and its formation through detected the tumor cells'imbalance growth and migration.Methods: To explore the formation of the"overlapping growth"of hepatoma cells through processing quantitative characterization on the growth of hepatoma cells in situ with morphological observation, Image Tool computer analyze system, statistical analysis, as well as the experimental methods of cell mechanics and biochemistry. (1) Observed the hepatoma cells overlapping layer formation process (2) Dissected the "overlapping growth" structure (3) Compared the difference of migration and adhesion force between hepatoma cells and hepatic cells.Result: (1)The ability of hepatoma cells to regulate cell morphological deformation was better than hepatic cells, the growth colony of heaptoma cells were symmetrical expansion and the "overlapping colony" in the center of growth colony ; (2) Using micropipette aspiration technique to suck the"overlapping growth plaque"of hepatoma cells, the"overlapping growth plaque"fell off from the substrate, leaving a blank area; (3)The adhesion strength of round cells which located in :overlapping colony"was lower than the spread cells;(4)Hepatoma cells'integrin expression was obvious more than hepatic cells; (5)Fn down-regulate the integrin express in hepatoma cells when cultured on the Fn coated surface, enhanced the cells'adhesion ability and morphological stability , but reduced the formation and aggregation of the round cells. (6)When the hepatoma cells in low density ,the ability of migration of spread cells was better than round cells ,however, when the cells growth to confluence, the round cells had active migration ability.7)Stretched the growth colony, the spread cells tended to migration to center.Conclusion: (1) The so-called overlapping growing area was actually consisted of many closely arrayed and piled round cells; (2) Round cells produced maybe caused by integrin abnormal expression and the effect on the hepatoma cells adhesion stability; (3)The formation of"overlapping growth colony"in hepatoma cells related to the regional distribution which caused by the difference of cell migration, adhesion between the spread cells and round cells in"overlapping colony";...
