Objective:To explore the protective effects of Rotundine injection on lung, liver and kidney damages after cerebral ischemia reperfusion in rats based on the changes of nitric oxide synthase (NOS) activities.Methods: The cerebral ischemia reperfusion models were established by Pulsinelli's.The rats were randomly divided into three groups: sham operation group (group S), ischemia-reperfusion group (group IR) and treatment group (group T) after reperfusion 2,6,12,24 and 48 hours. Group T received intraperitoneal dose of rotundine 20mg.kg-1 after occlusion, once every six hours. The rats of each group were decapitated at the varing time, collected samples of pulmonary, liver and kidney and measured the activities of NOS, blood creatinine (Cr) and alanine amino transferase(ALT).Results: (1) Compared with group S, the activities of total NOS were increased at 2h,12h and 24h in ischemia-reperfusion group (group IR), The activities of cNOS and iNOS were increased significantly at 2h and 12h separately.(2) Compared with group IR, The activities of eNOS increased much higher at 2h and the activities of iNOS were significantly decreased at 12h and 24h in group T.(3) There were no pathologic changes in group S, but in group IR, there were deferent pathologic changes and higher serum levels of ALT and Cr, especiallly at 12h(P<0.01). The pathologic changes were significantly improved in group T. Conclusions:(1) The various types of NOS play different roles in the pulmonary, liver and kidney damages in the different stages after brain ischemia-reperfusion in rats. Consitutive nitric oxide synthase protect the organs, but inducible nitric oxide lead to damages to the organs.(2) Rotundine injection can improve the damages through modulating the activities of different NOS. Rotundine can increases the activities of consitutive nitric oxide synthase in the early stage ang decreases the activities of inducible nitric oxide synthase in the late after brain ischemia-reperfusion.
