The Compound Erjingling polysaccharides was extracted and purified from Rhizome of King Solom Oseal and Fructus Lycii, it was the main function ingredients of Compound Erjingling. According to the report, the Erjingling polysaccharide can clear improve the content of hydroxyproline, the numbers of fibroblast and the activity of SOD in musculus skin and it can reduce the content of lipofuscin in organization of musculus skin, it have the effect to anti-aging. The Erjingling polysaccharide aslo have the effect to Alzheimer's dsease, it can stop the calcium accumulation in hippocampal cells, that can reduce the hippocampus cells damage caused by calcium overloading in hippocampus cells and it aslo can stop the hippocampus apoptosis caused by Glu induction effective.This paper mained focused on the extraction, isolation, purification and characterization of polysaccharide from Compound Erjingling were systematically studied.The main results were shown as follows:1 Extraction,isolation and purification of polysaccharide from Compound ErjinglingThe Erjingling polysaccharide(2JL) was came from the mixture of Rhizome of King and Fructus Lycii. It was by discolored,drew off monosaccharide,hot water extraction and ethyl alcohol precipitation.The Erjingling polysaccharide(2JL) was isolated and purified through DEAE- CelluLose(OH-) , to get six polysaccharide constituents. Constituent1 through Sepharose CL-4B and constituent4 through Sephadex G-200 gel permeation column chromatography , to get two polysaccharide components 2JL and 2JLⅡ.2 Physical,chemical and structural characterizations of Erjingling polysaccharideThrough HPLC detection and analysis proved that 2JLⅠand 2JLⅡwere homogeneous polysaccharides.Their neutral polysaccharide contents were 62.82% and 85.39%, protein contents were10.48% and 4.42%. The relative molecular weight of 2JLⅠwas 811000 and the relative molecular weight of 2JLⅡwas 2000 by GPC analysis. Through GC analysis proved that 2JLI was mainly composed of arabinose, xylose, galactose, mannose and glucose,their molratio was 0.141:0.019:0.189: 0.021: 0.213; 2JLⅡwas composed of galactose,glucose and mannose,their molratio was 0.082:0.283:0.190. This two polysaccharide components were glucoprotein complexes.By IR, periodic acid oxidize and MS analysis,2JLⅠand 2JLⅡmainly containα-glycosidic bond.Throughβ-elimination reaction analysis, the polysaccharides and amino acids in glucoprotein moleculars of 2JLⅡattended mode was O- connection. 2JLⅡboth can reaction with Congo red,this pointed that the structures 2JLⅡmaybe were helical structures.
