The Study On Ultrastructure Of Freeze-dried Spermatozoa

Objectives To investigate the ultrastructure differents of sperm between and after freeze-drying.Methods Healthy sperm was divided into five groups.Group 1 was a control group; ETBS was added in groupⅡ;ETBS and trehalose was added in groupⅢ;meanwhile,ETBS+trehalose+egg yolk was added in groupⅣ.GroupⅤwas freezed with Procedure freezing method..After four weeks,all five groups were observed by scanning electron microscope.Results The normal sperm ratio in five groups were(46.39±1.46)%,(55.75±1.29) %,(67.20±1.8)%,(63.16±2.63)%,(85.94±1.86)%,and the activity ratio of five groups were 70.71%±7.5%,21.74%±3.2%,32.15±5.19%,40.52%±2.86%, 35.12%±4.37%o p＜0.05Conclusions After freeze-drying,the sperm group with ETBS+trehalose+egg yolk had little ultrastructure damage.
