Objective To investigate the levels of microvessel density(MVD) , the expression of P-Akt,PTEN and vascular endothelial growth factor (VEGF) in human follicular thyroid carcinoma (FTC), and explore the correlation between them and the role of PI3K/Akt signal pathway in FTC.Methodsâ‘ MVD and expressions of P-Akt,PTEN and VEGF in 40 specimens of FTC tissues,10 specimens of thyroid adenoma tissues and 10 specimens of nodular goiter were detected by immunohistochemical PV6000 method.â‘¡The immunohistochemical results were analyzed using the software of SPSS11.5 by means of Chi-Square test,Rank sum test and Homogeneity test of variance.Resultsâ‘ The expression rate of P-Akt in FTC was higher than that of in non-FTC tissues.The result was of statistical significance (x~2=43.45,P<0.001),The expression rate and intensity of P-Akt in FTC were obviously elevated with the differentiation degree depressed(H=l2.119, P<0.01).â‘¡The positive rate of PTEN in nodular goiter,thyroid adenoma and FTC were 100% ,8 0% and 25%, the difference between nodular goiter and FTC, thyroid adenoma and FTC were of statistical significance(x~2=15.76,P<0.01;x~2=8.25,P<0.01),the difference between nodular goiter and thyroid adenoma was of no statistical significance(x~2=0.237 P > 0. 05).â‘¢The expression rate of VEGF in FTC and non-FTC was of statistical significance(x~2=20.00, P < 0. 001) ; the difference between nodular goiter and thyroid adenoma was of no statistical significance(P > 0.05).â‘£There were correlations between the MVD of FTC tissue and the expression of P-Akt and VEGF(P<0. 05); The MVD of P-Akt positive FTC tissue was higher than that of negative tissues(P<0.05);The MVD of VEGF positive FTC tissue was also higher than that of negative tissues(P<0.05);There were correlations between the expression of P-Akt and VEGF( x~2=4.17,P <0.05).Conclusionsâ‘ The hyperexpression of P-Akt play an important role in the secondary deposits and development of human follicular thyroid carcinoma.â‘¡The loss of expression of PTEN promote the invasion and metastasis of FTC through regulating the expression of P-Akt.â‘¢VEGF play an important role in the metastasis and development of FTC. Akt/VEGF pathway may be exist in FTC, Akt is the key factor in the pathway which regulated the expression of VEGF.
