OBJECTIVEStudy on the biologic activity of Seleno-polysaccharide of eucheuma. The main research is about anti-tumor and anti-viral.1) Study on the Seleno-polysaccharide of eucheuma impact on the rumor cell proliferation and induced apoptosis and Fas expression in vitro. The purpose is to understand selenium Eucheuma.2) Preliminary study of the antiviral effect of Seleno-polysaccharide of eucheuma and its impact on the absorption of the virus.METHODS1. The cultivation of normal and tumor cells.2. The inhibitory action to normal and tumor cells of Seleno-polysaccharide of eucheuma by MTT method.3. Flow cytometer was used to detect the change of the cell cycle, apoptosis and Fas by use of Seleno-polysaccharide of eucheuma.4. Cytoplasmic RNA from cell group and group of Seleno-polysaccharide of eucheuma was extracted by the Trizol method and mRNA levels of caspase-3, caspase-8 were assayed by reverse transcript-polymerase chain reaction(RT-PCR ) method respectively.5. The inhibitory action of virus by use of different concentration Seleno-polysaccharide of eucheuma by use MTT methods and study the effective of viral adsorption.6. All statistical analyses were performed using the software SPSS 13.0. RESULTS1. Seleno-polysaccharide of eucheuma has the inhibitory action to the tumor cells.2. FCM showed tumor cells in G1 phase decreased, cells in S and G2/M phase increased, the cell cycle was arrested in S and G2/M phase.3. The relative mRNA expression levels of caspase-3 and caspase-8 were significantly enhanced compared with normal controls4. Seleno-polysaccharide of eucheuma can inhibit the multiplication of virus. Adding Seleno-polysaccharide of eucheuma with virus at the same time can inhibit virus(P<0.05) better than adding it after virus having been adsorbed to cells (P>0.05) .CONCLUSION1. Seleno-polysaccharide of eucheuma had significant inhibitory effect on tumor with dose and time-effect relationship.2. Seleno-polysaccharide of eucheuma may induce Hela and HepG-2 cells apoptosis through arresting cells in S and G2/M phase.3. Seleno-polysaccharide of eucheuma may induce Hela and HepG-2 cells apoptosis through Fas pathway.4. The mechanism of anti-virus of Seleno-polysaccharide of eucheuma possibly arrested the adsorption of virus.
