Objective To prepare a desired specific immunoadsorbent for anti-AChR antibodies in myasthenia gravis patients. The adsorption of the prepared specific immunoadsorbent to anti-AChR antibodies in myasthenia gravis patients was studied by dynamic absorption test on the patients' serum. Methods 10% cellulose beads was synethesized by suspension regeneration. 20-40 sized cellulose beads as vector was activated by epichlorohydrin. The content of epoxy could achieve to 92μmol/g. As ligands, synthesized polypeptide of AChR at residues 67-76 were immobilized on cellulose beads to make immunoadsorbent. The adsorbents were reacted with patients' serum which contained anti-AChR antibodies at 37℃for 4 hours with circulation. Before and after the reaction, the optical densities of the antibodies were determined respectively and adsorption capacities were calculated. Results. The adsorbent with cellulose as carrier and polypeptide as ligand, was of high affinity with antibody in dynamic absorption test on the patients' serum. The adsorption capacity rose continuously as the time prolonged, and there was a peak after 2 hours of the adsorption. The adsorption capacity was 26.59%. Conclusion The adsorbent with cellulose as carrier and polypeptide as ligand, which is of high affinity with antibody, is an ideal immunoadsorbent in specific immunotherapy of myasthenia gravis.
