Aim:Studies the hot shock protein 27 through the RT-PCR law and the immunohistochemistry method to the spinal cord ischemia-reperfusion injury to pour into the different time interval gene and protein expression situation,further elaborated the hot shock protein 27 biologies characteristics in the protection mechanism of the spinal cord ischemia-reperfusion injury.Methods:The experiment to 2006-07/09 completes to the Fujian traditional Chinese medicine college laboratory,SD big mouse 48,are female and male does not limit,body weight 180-320 g,average 230 g,Stochastically divides into the normal comparison group and the spinal cord ischemia-reperfusion injury 0,1,2,4,8,12,24 h group(n=6),Uses Zivin and so on the method preparation the model of the spinal cord ischemia-reperfusion injury, After lacks blood 30 min to pour into the certain time again(0,1,2,4,8,12,24 h) Pours into fixedly with 4%paraformaldehyde,Excises the L3-4 double side centrum,fully revealed L3-4 hard membrane pouch,shuts off the peripheral nerve root,takes out the identical section spinal cord,Useing the RT-PCR reacting system and the immunohistochemistry method examines the HSP27 gene and protein expression of different groups and different time compartment.Results:48 SD big mouse enters the result analysis,①The HSP27 gene and protein expression elevates immediately after the spinal cord ischemia-reperfusion injury, respectively group of the spinal cord ischemia-reperfusion injury to have notable difference with the normal comparison group(P<0.05),②there was no significant difference between the groups of the 0,1 h,2h,12h,24h,③the HSP27 gene and protein expression of the spinal cord ischemia-reperfusion injury 4h and 8h are maximal and have notable difference with other sets(P<0.05),the HSP27 gene and protein expression has no notable difference between 4 h and 8 h group level(P>0.1). Conclusion:After the spinal cord ischemia-reperfusion injury the HSP27 gene and protein expression elevates immediately again,to 4h and 8h is the high point.Afterwards the HSP27 gene and protein expression passes along with the time reduces,but still had notable difference with the normal comparison group.
