Objective:To explore the mechanism of action of preserved amniotic membrane to inhibit neovascularization and inflammation after corneal alkaline burn.Methods:A total of 160 healthy mice were burned eyes with NaOH and divided into conventional treatment group(group A)and conventional treatment combined with amniotic membrane transplantation group(group B)randomly.The expression of VEGF,PEDF and NF-κB was detected on the 3rd,7th,10th and 14th day respectively by immunohistochemical SP method.Results:1.On the 3rd day,expression of VEGF decreased in group B compared to A (+:40.0%and 40.0%,++:10.0%and45.0%,+++:0%and5.0%,p<0.05).On the 7th day, they were 40.0%and 10.0%,30.0%and 35.0%,15.0%and50.0%,correspondingly(p<0.05).On the 3rd day,expression of NF-κB decreased in group B compared to A(+:55.0%and 20.0%,++:15.0%and45.0%,+++:10.0%and25.0%,p<0.05).On the 7th day,they were65.0% and40.0%,10.0%and30.0%,5.0%and20.0%,correspondingly(p<0.05).On the 10th and 14th day,expression of VEGF and NF-κB was no statistical difference in group A and group B(p>0.05).2.Expression of PEDF on the 3rd,7th,10th and 14th day in group B increased not remarkably compared to group A(p>0.05).4.There was positive correlation between the expression of NF-κB and VEGF in group A and B.Conclusions:1.Amniotic membrane transplantation maybe resist neovascularization and inflammation by inhibiting the expression of NF-κB and VEGF before the amoiotic membrane dissolved.2.Amniotic membrane transplantation can not increase the express -ion of PEDF in the corneal tissue.3.PEDF should be extracted from the amniotic me -mbrane for clinical effect of treatment.
