Objective The research was aimed to investigate the influence of TashinoneⅡA-Sulfoacid-Natrum(TSN) on the electrophysiology and pressure of rabbit's ventricle in vivo and the experimental arrhythmia.Material and methods1.The influence of TSN on the electrophysiology and pressure of rabbit's left ventricle in vivoForty New Zealand white rabbits were randomly assigned into five groups: control group, TSN 0.5mg·kg-1 group, TSN 1mg·kg-1 group, TSN 2mg·kg-1 group and TSN 4mg·kg-1 group, the electrode lead was inserted into ventricular wall and a tube was plugged into left ventricle, to observe and record the influence of different doses of TSN(0.5,1,2,4 mg·kg-1) on the electrophysiology and pressure of rabbit's left ventricle in vivo.2. The influence of TSN on experimental arrhythmiaStudy the influence of TSN on the experimental arrhythmic models, which were induced by barium chloride and chloroform-Adnephrin. Results low dose(0.5mg·kg-1 and 1mg·kg-1) of TSN had no obvious influence on the rabbit's left ventricular electrophysiology and pressure, large dose(2mg·kg-1 and 4mg·kg-1) of TSN could prolong left ventricular relative refractory period(LVRRP prolonged from 123.75ms±3.53ms and 123.12ms±3.72ms to 153.75ms±8.34ms and 157.54ms±4.62ms,P<0.05),left ventricular effective refractory period(LVERP prolonged from 119.37ms±4.17ms and 119.37ms±4.17ms to 152.5ms±7.55ms and 145.62ms±8.21ms,P<0.01 ),elevate ventricular fibrillation threshold ( VFT elevated from 2.81v±0.37v and 2.62v±0.51v to 5.68v±0.92v and 5.59v±0.94v,P<0.01), and could cut down left ventricular systolic pressure(LVSP was cut down from 108.62mmHg±2.06mmHg and 109.13mmHg±2.79mmHg to 102.37mmHg±2.67mmHg and 103.12mmHg±4.01mmHg,P<0.05),left ventricular end diastolic pressure(LVEDP was cut down from -4.12mmHg±3.52mmHg and -4.01mmHg±3.66mmHg to -7.50mmHg±4.2mmHg and -7.12mmHg±3.9mmHg,P<0.01) and the peak of intraventricular pressure change rate(+dp/dtmax was cut down from 5584.87mmHg/s±442.67mmHg/s and 5715.75mmHg/s±526.49mmHg/s to 5039.2mmHg/s±471.81mmHg/s and 5189.6mmHg/s±472.67 mmHg/s,P<0.05);and reduced the occurrence of arrhythmia of rats which was induced by barium chloride(the incidence reduced from 100% to 40%,P<0.05) , and decurtated the duration of arrhythmia of rabbits which was induced by chloroform-Adnephrin(the duration of arrhythmia decurtated from 296.01s±47.35s to 265.37s±38.85s,P<0.01).Conclusions TSN may have the efficacy of increasing electrical stability in cardiac muscle and reducing the occurrence of arrhythmia.
