Study On Therapy Of Wu Zi Yan Zong Tang To Leber Hereditary Optic Neuropathy And Effects To Mitochondrial Membrane Potential

Objective1. Through testing the visual acuity,visual field and VEP of the patients with Leber hereditary optic neuropathy (LHON) before and after treatment, study the effects of wu zi van zong tang to clinical expressions of patients with LHON.2. Through determining mitochondrial membrane potential of the patients with LHON before and after treatment, study the effects of wu zi yan zong tang to the function of mitochondrial of patients with LHON.3. Wish of finding a method to cure LHON, and try to illuminate the mechanism of the method.MethodWe separated the 30 patients of LHON consistent with our diagnostic criteria into two groups: The 20 patients of the treatment group take the wu zi yan zong tang and coenzyme Q₁₀. The 10 patients of the control group take coenzyme Q₁₀. The period of clinical trial was 3 months. Before and after the clinical trial we tsted the visual acuity,visual field and VEP,tested the mitochondrial membrane potential through the technique of flow cytometry of the 30 patients.Result1. Of the patients in the treatment group, visual acuity was greatly improved, and the mean sensitivity was improved, the mean defect of visual field reduced, the amplitude of VEP was elevated significantly after 3 months' treatment. The mitochondrial membrane potential of peripheral blood lymphocytes was improved.2. Of the patients in the control group, visual acuity was improved, but the mean sensitivity was not improved, the mean defect of visual field didn't reduce, the amplitude of P-VEP was not elevated after 3 months' treatment. The mitochondrial membrane potential of peripheral blood lymphocytes was not improved.3. Compared with the pure western methods, using the cooperation of Chinese and Western medicine method to take wu zi yan zong tang and the coenzyme Q₁₀ may improve the LHON patient's visual acuity; improve the visual sensitivity, thus improve visual function; elevate the amplitude of visual evoked potential P100 wave thus improve optic nerve conduction function; enhance the mitochondrial membrane potential of peripheral blood lymphocytes, thus to improve the function of mitochondrial.4. The Chinese medicine general symptom, such as lumbago or spermatorrhea was improved of the LHON patients in the treatment group.5. With the method of linear correlation and regression analysis, we found the mitochondrial membrane potential of peripheral blood lymphocytes and the vision, mean sensitivity, the latency and amplitude of pattern- visual evoked potential P100 wave of the patients with LHON have linear regression relations, the regression contributions are 42%, 27%, 31%, 16% respectively.Conclusion1. Using the cooperation of Chinese and Western medicine method to take wu zi yan zong tang and the coenzyme Q₁₀ may improve the LHON patient's visual acuity and visual sensitivity, thus improve visual function; elevate the amplitude of pattern-visual evoked potential P100 wave, thus improve optic nerve conduction function; enhance the mitochondrial membrane potential of peripheral blood lymphocytes, thus to improve the function of mitochondrial.2. Using the pure Western medicine method to take coenzyme Q₁₀ can't improve the LHON patient's visual acuity and visual sensitivity, can't elevate the amplitude of pattern-visual evoked potential P100 wave. So it can't improve visual function or improve optic nerve conduction function. It can't enhance the mitochondrial membrane potential of peripheral blood lymphocytes with this method. So it can't improve the function of mitochondrial.3. Compared with the pure western methods, using the cooperation of Chinese and Western medicine method to take wu zi yan zong tang and the coenzyme Q₁₀ may improve the LHON patient's visual acuity and visual sensitivity, thus improve visual function; elevate the amplitude of visual evoked potential P100 wave, thus improve optic nerve conduction function; enhance the mitochondrial membrane potential of peripheral blood lymphocytes, thus improve the function of mitochondrial.4. The Chinese medicine general symptom, such as lumbago or spermatorrhea may be improved of the LHON patients with the cooperation of Chinese and Western medicine method to take wu zi yan zong tang and the coenzyme Q₁₀. It may have the function of tonifing kidney, tonifing essence and improving eyesight.5. The mitochondrial membrane potential of peripheral blood lymphocytes and the vision, mean sensitivity, the latency and amplitude of pattern- visual evoked potential P100 wave of the patients with LHON may have linear regression relations.
