The Study About Liweininglu Influences The Senile And Atrophic Genital Tract And Vaginal ER-β Of OXV SD-rat

Objective:To better know the effect of Liweininglu,which includs anti-aging and adjusting the internal environment of vagina of ovariectomized(OVX) Sprague-Dawley(SD) rat.Study how the acid and sweet herb(such as Liweininglu) educes enriching and adducting function.Study the mechanism of Liweininglu about improving the senile and atrophic vagina.To research how the Liweininglu adjusts the level of the ER-β.Give the basic study of the therapy about atrophic vaginitis.Methods:Divide the 80 female SD-rat into six groups:sham operated group(J), model group(M),blank group(K),positive control group(B),high dose group(G),low dose group(G).After the operation,continue vaginal smear, according to the preliminary experiment,bring the animal for 20 days, the organs of the model can be as senile as the old ones.In the 21^st day start to administration.Measure the pH,observe the secretion and the state of mucosa of vagina. Execute the animals,calculating the index number of uterus and vagina, then preparing the tissue homogenate of uterus and vagina to measure the SOD and MDA.The paraffin sections of uterus and vagina were used to HE staining,staining for vaginal glucogen,staining for elastic fibers and using the immunohistochemistry way to measure the ER-βin vaginal tissue.Results:After OXV 20 days,the models with ageing genital tract,have high pH in vagina,high weight,low mark of vaginal heath,low index number of uterus and vagina,low SOD,high MDA,the phlegmonosis of tissue,glucogen and elastic fiber are lower,having significant difference with sham operation(P＜0.05).Liweininglu can depress the pH of the vagina,control the weight,and raise the mark of vaginal heath,raise index number of uterus and vagina,raise SOD.Also can low down MDA,change the state of the tissue of uterus and vagina,increase glucogen and elastic fiber,and suppress ER-βexpressing,having significant difference with model group (P＜0.05).Liweininglu low dose group raises SOD better than high dose group,but depress MDA worse than high dose group.There are no significant difference between the two groups(P＞0.05).Conclusion:The Liweininglu can degrade the vaginal's pH of OVX rat,improve the ageing and shrinking gential tract,raise elasticity of vaginal tissue and suppress the increase of ER-β.And have no obviously differences with Premarin.The blank only can moist the illness vagina without curing effects.The index number of uterus and vagina reflects the estrogen-like effect of medicine.So the Premarin shows the strong estrogen-like effect than other groups,and makes the endometria thickening obviously(P＜0.05). Using estrogen longtime,having the dangerous of cancer.The Liweininglu is made of Chinese herb,which provides a safety way for treating atrophic vagina.At the same time,sham operated group,blank group and administration team have lower ER-βlevel than the model does.May be low pH can suppress ER-βexpressing.
