Effects And Mechanisms Of Lanyuzan Granula On DMN-induced Hepatic Fibrosis In Rats

AIM: Hepatic fibrosis (HF) is a kind of serious hepatic diseases and is the common final of many chronic hepatic diseases. However the mechanism is still unclear to us and there is no satisfactory drug therapy for it. Glucocorticoids is the common choice in treating PF nowadays,yet many studies indicated that it could induce some serious diseases such as osteoporosis and et al. Traditional Chinese medicine has a bright future in preventing and treating HF and many have made great progress in clinical therapy. In this study, we interfered the process of fibrosis with a Chinese medicine Lanyuzan granula to study its protect effect on early experimental hepatic fibrosis and explored the potential mechanism in DMN-induced rats'HF,then settle a theoretical foundation for its clinical use.METHODS: (1) 30 SD rats were randomly divided into Control groups, Model group, Lanyuzan groups (dose:4g/kg) , predniosone groups lanyuzan+ predniosone groups, and were given one intraperitoneal injection of 2mg/kg of DMN(5 mg/L) in 0.01mL 0.9% saline or an equivalent volume of 0.9% saline to set a model of hepatic fibrosis. On day 1,each HP group was given Lanyuzan abstract,predniosone respectively except DMN model group. On day 28, rats in each group were sacrificed and livers were harvested. The right livers were fixed in 10% formaldehyde for 1 day, paraffin-embedded and sliced for histopathological studies (HE staining and MASSON staining),the left lungs were homogenized for biochemical assays of SOD activity and MDA, GSH, MAOcontent. (2) 42 SD rats were randomly divided into Control groups, Model group, Lanyuzan granula high groups (dose:4g/kg), Lanyuzan granula middle groups(dose:2g/kg) and Lanyuzan granula low groups(dose:1g/kg) , predniosone groups, and were given one intraperitoneal injection of 2mg/kg of DMN(5 mg/L) in 0.01mL 0.9% saline or an equivalent volume of 0.9% saline to set a model of hepatic fibrosis. On day 1,each HP group was given Lanyuzan granula,predniosone respectively except DMN model group. On day 28, rats in each group were sacrificed and livers were harvested. The right livers were fixed in 10% formaldehyde for 1 day, paraffin-embedded and sliced for histopathological studies (HE staining and MASSON staining),the left lungs were homogenized for biochemical assays of SOD activity and MDA, GSH, MAOcontent.RESULT: Compared to model group,fibrous degeneration were alleviated in Lanyuzan granula group significantly; pathological grading of fibrous degeneration show that fibrosis were remarkable alleviated in Lanyuzan granula group and predniosone group(P<0.01),Ⅰ~Ⅱ-rate HF took a dominant position in Lanyuzan granula group,Ⅱ~Ⅲ-rate HF were displayed in predniosone group.MDA,MAO contents of homogenized lungs in Lanyuzan granula group lowed (P<0.05), GSH contents of homogenized lungs in Lanyuzan granula group and predniosone group increased (P<0.05);CONCLUSION: The results of our experiments showed that Lanyuzan granula and Prednisolone could retard and restrain HF in DMN-induced rats. Lanyuzan granula could decrease TNF-αcontent in homogenate of lung, heighten the activity of SOD, lower damnification of lipid peroxide(LPO), enhance the power of eliminating and restrain Oxygen radical, alleviate inflammation and decrease phlogistic cells,So effect on early experimental pulmonary fibrosis. The possible mechanisms of Lanyuzan granula preventing and treating HF is improve the oxidation/deoxidation balance and restrain inflammation. and alleviate and restrain pulmonary fibrosis.
