Objective: To research the protection of the allitridi on the brain functional lesion in diabetes mellitus rats with High homocysteine, and to observe the effect of allitridi rivalry the damaging effect of homocysteine to endotheliocyte and to find out the mechanism of action.Methods: 1. 60 wistar male rats were fed with animal feeds for 1 week, and ten rats were pulled out randomly as the normal group, the rest 50 rats were injected with streptozotocin (STZ) into abdominal cavity for diabetes mellitus animal model. Choosing 40 from the rats according to qualification were divided into four groups, a block group(10 rats), a model control group (10 rats), a western treatment group (10 rats), a allitridi treatment group (10 rats) and the surplus 10 rats as a normal control group. All rats of the allitridi treatment group,the western treatment group and the model control group were fed with high methionine forag for preparing diabetes mellitus rats with high homocysteine, the normal group and the block group were fed with common forage. The allitridi treatment group were injected with allitridi 8mg/kg into abdominal cavity every day. The western treatment group were given powder, including folic acid(0. 45 mg/kg), vitB6 (2.7 mg/kg), and vitB12 (6. 75μg/kg) every day. After giving medicine for 6 weeks, the BAEP, electric maze and transcranial doppler sonography (TCD) were detected. The fasting blood sugar (FBG) , blood biochemistry insulin, homocysteine of serum, FINS, SOD, MDA and monoamine neurotransmitter were determined in all rats. To observe histomorphology of seahorse and to determine the target of molecular biology of seahorse . 2. We divide the cultivate human allantoic veins endotheliocyte into high homocysteine group (normal control group), allitridi group with flushing dose , allitridi group with middle dose, allitridi group with low dose, the block group , and taking out of supernate fluid for determining the level of ET, NO, SOD, MDA, LDH and observing survival rate of the cell with MTT chromatometry at the time of 12h,24h,36h after giving the drug .Results: 1.The interspike interval of BAEPⅠ-ⅢIPL andⅠ-ⅤIPL waves of the allitridi treatment group were significantly lower than that of the blank groups, the model control group (P<0. 05). It hint that allitridi can improve central conduction time of diabetes mellitus rats with high homocysteine.2. The capacity of learning and memory result of the allitridi treatment group surpassed significantly than that of the blank group/the model control group (P<0. 05-0. 01). Allitridi can improve the damage of the capacity of learning and memory caused of diabetes mellitus rats with high homocysteine, but western medicine can not.3. Comparing with the blank group/the model control group,it is statistical significance of the blood flow rate and CVR between the allitridi treatment group and the blank group/the model control group (P<0. 05), but these of the allitridi treatment group were similar to that of the normal control group(P>0. 05) , the effect of the group was not apparently statistics difference with the western treatment group (P>0. 05) . Allitridi also have the effect of improving cerebral blood flow of diabetes mellitus rats with high homocysteine.4. Comparing with the blank group/the model control group , allitridi also can raise ISI and improve IR obviously than the two groups. It is a statistics difference (P<0. 05). After treating, blood sugar of allitridi treatment group was lowering than the blank group/the model control group,it is a statistics difference (P<0. 01), and the arlicin treatment group surpassed obviously than the western treatment group (P<0.05). Allitridi also have the effect of raising ISI, improving IR and lowering blood sugar.5. The level of Hcy of the allitridi treatment group descended obviously than the model control group (P<0. 01) , but there have no statistics difference between allitridi treatment group and western treatment group (P>0. 05). Allitridi have the effect of lowering the level of homocysteine.6. After giving allitridi, the active of SOD was higher than the normal group (P<0. 05) . The level of MDA was lower than the blank group/the model controlgroup(P<0. 05-0. 01), but it was similar to the normal group(P>0. 05). Allitridi have the effect of cutting down the level of MDA and heightening the active of SOD.7. After giving allitridi, the content of NE, DA of brain tissue was obviously lower than the model control group (P<0. 05-0. 01) . The level of 5-HT is higher than the western treatment group.The express of NOS and Ach E albumen was obviously higher than the blank group/the model control group (P<0. 01) , and the effect surpass the western treatment group (P<0. 05-0. 01) . Allitridi have the effect of regulating synthesis of monoamine neurotransmitter, the level of NOS and improving the active of Ach E.8. The express of IGF-I heighten obviously than the blank group/the model control group (P<0. 05). Allitridi have the effect of up-regulating the express of nerve growth factor.9. Comparing with the blank group/the model control group, the express of Bax of allitridi treatment group was obviously down regulation, but the ratio of Bcl-2/Bax and the count of hippocampus tissue was obviously up regulation. There is statistical significance (P<0. 01) . The effect surpass the western treatment group (P<0. 01) . Allitridi have the effect of suppressing apoptosis of hippocampus tissue.10. In the endotheliocyte culture experiment, the level of ET of allitridi group with middle dose,low dose was obviously lower than the model control group but the level of NO heighten obviously(P<0. 05). Allitridi can promote endotheliocyte synthesis , release NO and lower the level of ET.11. In the endotheliocyte culture experiment, the level of MDA of allitridi group with the middle dose/the low dose descend degrade obviously, and the activity of SOD heighten obviously. There is statistics difference (P<0. 05-0.01) . The level of LDH of allitridi group with middle dose, low dose was obviously lower than the model control group (P<0. 01). The value of A of photoabsorption was similar to the blank group but heighten obviously (P<0. 01). Allitridi also can degrade the level of MDA and step up the activity of SOD and endotheliocyte and lower the level of LDH.Conclusion: Allitridi can markedly cut down blood sugar and the level of homocysteine. It also may elevate and improve the brain functional lesion in diabetes mellitus rats with high homocysteine by many kinds of paths, including of to amend sensitivity of insulin, to lower insulin resistance , to improve cerebral blood flow, to resist free radical, to regulate neurotransmitter, to increase expression of nerve growth factor and suppress apoptosis of hippocampus nerve cell.
