Objective To investigate detecting Cyclin D1 protein expression by MaxvisionTM and detecting Cyclin D1 mRNA by reverse transcriptase polymerase chain reaction (RT-PCR) for its application to diagnostic and differential diagnostic price of mantle cell lymphoma (MCL).Methods Paraffin-embedded samples of 15 cases of MCL, 7 cases of B small lymphocytic lymphoma,6 cases of follicular lymphoma, 10 case of lymphoid reactive hyperplasia and 3 cases of nodal marginal zone B-cell lymphoma as control group were retrieved from archival materials. Cyclin D1 protein was detected by MaxVisionTM and RNA was extracted form paraffin-embedded samples by RNA extracting kit,then RT-PCR,production analyzed by 1% agarose gel electrophoresis. House-keeping geneβ-actin was choosen as internal control for detecting RNA.Resultsβ-actin mRNA expression was detected 37 cases of all 41 cases (90.2%).Cyclin D1 protein was expressed in 10 of the 15 MCL (66.7% ).No Cyclin D1 expression was found in the control group. Cyclin D1 mRNA was detected 13 (86.7%) andβ-actin mRNA detected 14 (93.3%) in nodal MCL of 15 cases by RT-PCR in paraffin-embedded tissues. The positive rate of Cyclin D1 mRNA was 92.2% (13/14) in mantle cell lymphomas after exclusion of the 1 cases which were negative for both Cyclin D1 mRNA andβ-actin mRNA. Cyclin D1 mRNA was not detected in other nodal small B-cell lymphomas or lymphoid reactive hyperplasia, except 1 case of B small lymphocytic lymphoma. The positive Cyclin D1 protein expression and Cyclin D1 mRNA added up to 14 cases ,accounting to 93.3%(14/15).Conclusions Cyclin D1 protein expression was detected by immunohistochemical method and Cyclin D1 mRNA detected by RT-PCR had higher specificity and feasibility,which were helpful for optimization practice correction to diagnosis and differential diagnosis of mantle cell lymphoma in paraffin-embedded blocks by combination.
