Objective To investigate the protective effect of Edaravone( MCI-186) on oxidative injure of endothelial cells induced by H2O2;to examine the effect of MCI-186 on the expression of ICAM-1mRNA and MCP-1 mRNA and to study the effects of MCI-186 in atherosclerosis,the possible mechanism of its antiatherosclerosis.Methods In order to gain the oxidative injure cell model,we treated endothelial cells with H2O2and observed the oxidative injure of cells after treated by the different concentrations H2O2 as well as after treated by the same concentration H2O2 for different times;Endothelial cells were incubated with MCI-186 and H2O2 and then the protective effect of different concentrations MCI-186 on the oxidative injure of endothelial cells was elucidated,the expression ofICAM-1 mRNA and MCP-1 mRNA was observed;the cell growth condition was investigated with inverted microscope; the viability of cells was determined by MTT assay;the level of cells lipid peroxidation was detected by measuring MDA ;the expression of ICAM-1mRNA and MCP-1 mRNA was measured by RT-PCR.Results MCI-186 can decrease the extent of contracted endothelial cells damaged by H2O2; MCI-186 can diminish the level of MDA that was produced because of the oxidative injure of endothelial cells; the viability of EC can be improved by MCI-186; RT-PCR showed that ICAM-1 and MCP-1mRNA was expressed in all groups.Higher level of ICAM-1 and MCP-1mRNA expression induced by H2O2 was observed in H2O2 Stimulated group compared with that in control group (P<0.05).After being treated with MCI-186,The expression of ICAM-1 and MCP-1mRNA in H2O2+ MCI-186 group was reduced (P<0.05 ).Conclusions (1) The ICAM-1 and MCP-1mRNA incultured endothelial cells is markedly induced by H2O2(2)MCI-186 could decrease the oxidative injure of endothelial cells induced by H2O2;(3) MCI-186 could inhibit the expressions of ICAM-1 and MCP-1 mRNA induced by H2O2 .That may be one of the mechanisms of antiatherosclerotic effect of MCI-186.
