Objective: To exam ine the effect of telmisartan (TEL) on p revent ing the injury of human umbilical vein endothelial cells (HUVEC) induced by high glucose (HG ) and the change of product ion of peroxisome proliferator activated receptorsγ(PPAR-γ) . Investigate protective effect mechanism of telmisartan on endothelial cells exposed to high glucose .Methods: HUVECs was cultured and divided into three groups: HG group(33.3mmol/L),HG+TEL group (TEL: 1×10-8mol/L,1×10-7mol/L and 1×10-6mol/L) and blank control group. The changes of growth inhibition rates were monitored by MTT at an interval of 2,12, 24, 36 and 48 hours.HUVECs was divided into blank control, HG, HG+TEL(10-6mol/L) and TEL control group renew. Collecting supernatant at 2,12, 24, 36 and 48 hours. The total activity of SOD by xanthine oxidase test, volume of MDA was measured by thiobarbituric acid test,,that of NO by nitrate reductase and ET by radioactivity immunifaction.Western blot analysis technique was used to determined the protein expression of peroxisome proliferator activated receptorsγ(PPAR-γ) in HUVECs 24h after treatments.Result: 1. MTT: With the increased length of existence of high glucose, the growth inhibit rates increased accordingly. Contrast to HG group, TEL(1×10-6mol/L ) decreased the growth inhibit rates at12h, and TEL(1×10-8mol/L, 1×10-7mol/L) at the 48h. It is showed that TEL can reduce the effect of HG significantly (P<0.05). 2. SOD: At every culture moment(except 2h), SOD of HG group is lower than control, that of HG+TEL group higher than HG group. 3. MDA: At every culture moment(except 2h), MDA of HG group is higher than control, that of HG+TEL group lower than HG group. 4. NO, ET: At every culture moment(except 2h), NO of HG group is lower than control, that of HG+TEL group higher than HG group. The change of ET was contraty. 5. PPAR-γexpression: At 24h, PPAR-γexpression of HG group is lower than control, that of HG+TEL group higher than HG group.Conclusion: The high glucose inhibite the growth and functions of HUVEC. Telmisartan protect the endothelial function exposed to high Glucose levels, block oxidative stress and increases the expression of in endothelial cell disrupts by high glucose.
