The incidence of obesity has obviously increased with the development of economy and the level of human life.Recently ,many studies have shown that obesity is highly related with insulin resistance(IR).Many studies showed that adipose tissue plays an important role in the mechanism of IR by excreting some key cytokines such as tumor necrosis factor-α(TNF-α), interleukin-6(IL6) and so on,but not only acting as the main organ depoting energy. Futhermore, Increasing evidence from human population studies and animal research has established correlative as well as causative links between chronic inflammation and insulin resistance.Some adipocytokines such as TNF-α,CRP,interleukin-6 are all related with IR.They also has great correlationship with the development of type 2 diabetes mellitus and its chronic vascular complications .Furthermore,a great deal of clinic observations showed that complex Salvia miltiorrhiza drill could improve the retinopathy , peripheral neuropathy and renal lesions related with type 2 diabetes.But there are still no reports about whether complex Salvia miltiorrhiza drill could affect the innate inflammation state in the diabetes patients or experimental models.So,the objective of this study is to research the effect of complex Salvia miltiorrhiza drill on the serum TNF-α,IL-6 in the type 2 DM rats and to probe the mechanism of complex Salvia miltiorrhiza drill acting on the diabetic complications,hoping to provide some new messages and theories for the use of complex Salvia miltiorrhiza drill in the cure of diabetes. Objective: To investigate the the effect of complex Salvia miltiorrhiza drill on the improvement of inflammatory mediators such as TNF-αand IL-6 of type 2 diabetes mellitus in the rats.Research design and methods: Raising all the rats in the environmentally controlled conditions with two different kinds of diet.The experimental group containing 40 rats were raised by the high-glucose and high-fat diet ,while its 10 counterparts were given the standard chow food.Grouping those whose weight were 20 percentage more than that of the mean level of the control group into diet-induced obesity rats.And then copied the type 2 diabetes mellitus models by intraperitoneal injection of 30 miligram per kilogram (of the weight of rats ) with 1% streptozotocin.Dividing the diabetic rats into four groups :diabetic rats with drinking water, diabetic rats treated by intragastric administration of complex Salvia miltiorrhiza drill in the dose of 50,150,450 milligram per kilogram (of the weight of rat ) per day for 4 weeks .Weighting all the rats and measureing their body length and and fasting blood glucose every two weeks. While blood samples were obtained from the orbit veins with fasting abdomen in the morning after 6 weeks'raising ,when diabetes mellitus models were successflly made and after 4 weeks'complex Salvia miltiorrhiza drill intervention . Centrifuged the blood samples and kept the serum samples under -80℃to measure total cholesterol (TC ),triglyeride (TG ),low-density lipoprotein(LDL),high-density lipoprotein (HDL),tumor necrosis factor-α(TNF-α),interleukin 6 (IL-6),insulin levels and so on .All the data were expressed in Mean±SD,and analysed with the analysis of variance by SPSS 13.0 for windows.Assays: The levels of serum TNF-α, IL-6 and insulin were measured by radioimmumoassay method.The outcomes of blood fat were gotten from automatic biochemistry analysing machine.The fasting blood glucose were measured by glucometer Surestep.Evaluation standard: Based on the body weight ,those fed by the high fat diet had 20 percentage more than the ones in control group were referred as obesity rats.The fasting blood glucose(FBG)≥7.0mmol/L or the random blood glucose(RBG)≥13.9 mmol/L were included into type 2 diabetes mellitus subjects .Results:The level of weight ,lee's index number,fasting blood glucose ,TC,TG,LDL,INS,TNF-α,IL-6 of dietary induced obesity rats were statistically higher than that of control ones.But the level of HDL was obviously lower than that of control group. The differences were statistically significant at the 0.05 level.After treated by intragastric administration of complex Salvia miltiorrhiza drill for 4 weeks,the level of TC,TG,LDL,TNF-α,IL-6,weight and lee's index number decreased and that of fasting serum HDL elevated in the 450mg/kg?d group.What's more,the levels of TC,TG,LDL,TNF-α,IL-6 and FBG were statistically lower ,whil the serum HDL was much higher ,than that of diabetes group administered with drink water and that of 50mg/kg?d group. Though the levels of TG,HDL,IL-6,FBG,and INS didn't have statistical difference,levels of TC,LDL,TNF-αin the 150mg/kg?d group were obviously higher ,compared with that of 450mg/kg?d group,Conclusion: 1.High-glucose and high-fat diet could induce obesity,hyperlipodemia,hyperinsulinism and hyperglycemia in tha rats ,causing the development of insulin resistance.2.Treating the diabetic rats with complex Salvia miltiorrhiza drill could improve the lipid metabolism,glucagon hemostasis and inflammatory situation effectively.And the effects of this drug parallel with the administration of dosage.
