| Objective: The aim of this study is to investigate the biological significance of HIF-1α,VEGF,EphB4 and its ligand ephrinB2,MVD in human brain astrocytomas,and to explore the relationship among the different tumor grades,through observing their protein expression.Method: Using tissue chip technique to create the tissue chips,which consist of 60 tumor cases and 13 controlled normal brian tissues specimen , and combining with MaxvisionTM immunohistochemistry method ,We detected the expressions of HIF-1α,VEGF and EphB4/ephrinB2 of each experimental agent ,then caculated its mircrovessel density(MVD) by observing CD34 protein expression in endothelial cells in chips。Result: Expression of each experimental agent in 60 cases of human brain astrocytomas as follow (the positive cases including those hadro-positive ones): Expressions of HIF-1αwere positive in 35 cases,expressions of VEGF were positive in 38 cases ,expressions of EphB4 were positive in 40 cases ,and expressions of ephrinB2 were positive in 44 cases。The expressional ratio of HIF-1α,VEGF and EphB4/ephrinB2 was 58.3%,63.3%,66.7% and 73.3% respectively。Expression of each experimental agent in 13 cases of controlled normal brain tissues as follow:Expression of HIF-1αwas positive in zero case,expression of VEGF was positive in zero case,expression of EphB4 was positive in zero case ,and expression of ephrinB2 was positive in one case。The expression ratio of VEGF,EphB4,HIF-1α,and ephrinB2 was 0%,0%,0% and 7.7% respectively。The distinction of expression of HIF-1α,VEGF,EphB4/ephrinB2 between human astrocytomas and controlled normal human brain tissues are significant(P<0.05)。The low degree astrocytomas consisted of 32 cases,and the high degree astrocytomas consisted of 28 cases。In former group the expressional ratio of HIF-1α,VEGF and EphB4/ephrinB2 was 40.6%,46.9%,46.9%and 54.4% respectively;in the later group the corresponding expressional ratio of them was 78.6%,82.1%,89.3% and 89.3%。There were significant deviation between them(P<0.05)。The average MVD value of 60 astrocytoma cases was 43.48±14.1,the average MVD value of 13 controlled normal human brain tissues was 11.34±9.9,the MVD difference between the two groups was bovious(P<0.05)。With tumor grade rising the MVD became higher。There was significant difference between the low degree group and the high degree one。The expression of HIF-1α,VEGF,EphB4/ephrinB2 and the MVDs were significantly related to tumor histopathological grade(P<0.05),but not related with gender,age,tumor size and localizaton(P>0.05)。Conclusion:Over expression of HIF-1α,VEGF,EphB4/ephrinB2 and the average MVD values are closely associated with tumor histopathological grade。They may be used as markers of malignant progression and poor prognosis in patients with astrocytoma。... |
