Objective: To observe the effects of preventive moxibustion on "Nei Guan"and "Tan Zhong" on expression of heat shock protein 70 and on the general structure histomorphology, to reveal the protective mechanism of preventive moxibustion on rat myocardial ischemia-reperfusion injury, then provide scientific experiment evidence.Method: Using rat ischemia-reperfusion injury model in vivo, thirty male Wistar rats were randomly divided into five groups .Purely Ischemia-reperfusion group: the chest was opened and left anterior coronary was occluded by suture silk ,the occlusion was maintained for 20 minutes and released with reperfusion for 40 minutes ;Ischemic preconditioning Group (IP) : prior to ischemic reperfusion ,three cycles of 5 minutes of myocardial ischemia follow by 5 minutes of myocardial reperfusion , after which the procedures were the same to ditto; Preventive moxibustion Group(PM): 7 days prior to ischemic reperfusion ,moxibustion was given on "Nei Guan" "Tan Zhong" ,30 minutes twice daily .After which the procedures were the same to ditto .Sham Operating Group : the chest was opened and left anterior coronary was exposed with no further procedures; Control Group: Cadiocyte HE staining to observe change of histomorphology, Ultra structural pathological changes were observed with light microscope and the expression of heat shock protein 70 associated with apoptosis was analyzed by immunohistochemistry technique.Results : Preventive Moxibustion Group (PM) and Ischemic preconditioning Group (IP) was notable diversity compare with Purely Ischemia-reperfusion group in every term of index. Preventive Moxibustion Group (PM) and Ischemic preconditioning Group (IP) were notable diversity compare with Sham Operating Group and Control Group. Preventive Moxibustion Group (PM) and Ischemic preconditioning Group (IP) was not notable diversity. Conclusion: Preventive moxibustion have effects on cardiac muscle in myocardial ischemia rats, Preventive moxibusion can promote expression of heat shock protein 70, and Preventive moxibusion can induce the tolerance of heart muscle ischemia as a pretreatment method and relieve the impairment of cardial muscle after ischemic reperfusion. |
