Effect Of Neizhang Pill On Adolescent Polycystic Ovary Rats

Objective:The aim of this study was to investigate the effects of Neizhang Pill in rats with polycystic ovary and to explore the possible mechanism of Neizhang Pill in cells and molecules levels.Ovarian morphology,serum hormone,apoptosis of granulosa cells were observed with optic microscope,immunohistochemical staining and terminal deoxynucleotidy transferase-mediated dUTP nick end labeling(TUNEL).Expect the effect mechanism in cell and molecules levels posted by Neizhang Pill.Material and method:24-day-old immature female SD rats(n=60)were separated into two groups at random.One group was PCOS animal model(n=50),the other group was used as control(n=10).Progesterone-synchronized ira-mature rats were used as the animal model for Polycystic ovary(PCO)by injecting 1.5 IU hCG twince daily to 36 days old.And then separated 3 groups(10 rats per group)the model group treated with Neizhang Pill for 30 days,separated 10 rats from the model group as the control group.After treatment,killed the rats.Blood specimens were centrifuged at 3000r/min for 20 minutes,the supernatant was collected for measuring E₂,T,LEP and IGF-1.Weighed the ovarian weight,measured the thickness of thecal cells and granulosa cells in general HE staining section.The expressions of apoptosis in granulose cells by TUNEL labeling.Results:There were no difference of body weight between Neizang Pill treatment groups and control group before pouring Neizhang Pill.But after pouring,the treatmentgroups significantly lightened.In PCO rats the number of Iuteum decreased but the number of cystic follicles increased contrarily.The cellular layer of ovary granulosa cell of the rat with polycystic Ovary got thinner and thecal cell became thicker.BSHXT had the effect of increasing the cellular layer of ovary granulosa cell and decreasing the cellular layer of thecal cell for the rats with polycystic ovary.In PCO rats,serum T,LEP and IGF-1 levels were higher and in the treatment groups serum T and LEP levels significantly lower than the control group.Serum E₂ levels were higher than the control group.The treatment groups' expressions of apoptosis in granulose cells differ from the model group's. Conclusions:This study set up PCO rat modle,the changement of ovarian pathology and incretion are similar as the patient of PCOS.Neizhang Pill could effect on PCO rats' ovarian morphology and by this way to improve ovarian function.Neizhang Pill could improve internal secretion by reduce the levels of T,LEP and IGF-1.Neizhang Pill could promote ovulation by inhibit ovary granular cell apoptosis.
