| Objective To investigate the effects of Tetramethylpyrazine(TMP) on hepatic stellate cells(HSC-T6) about its cell proliferation,collagen expression and some related cytokines. As an exploratory study to investigate the effect of TMP on anti-hepatic fibrosis.Methods HSC-T6 were cultured with varying doses of drugs. The proliferation of HSC-T6 was measured by MTT assay. Collagen and hyaluronic acid(HA) were measured by ELISA assay. MMP13 mRNA and TIMP1mRNA were measured by RT-PCR. CTGF were measured by western blot.Results In some range,TMP could inhibit the proliferation of HSC-T6 and induce the apoptosis in a dose-dependent manne(rP<0.05).It could decrease the expression of CTGF,it could decrease the expression collagen(type I and type III) and HA also(P<0.05).And their descent shows a positive correlation (r:0.861,0.807,0.868,P<0.01).TMP can improve the expression of MMP13 mRNA (P<0.05),make ratio increation of MMP13/TIMP-1 ,to promote degration of collagen.Conclusion TMP have effect of anti-hepatic fibrosis. The first possible mechanism may be that TMP can inhibit the proliferation of HSC-T6 and induce the apoptosis in a dose-dependent manner.The second possible mechanism may be that TMP can inhibite the expression of collagen by the means of inhibition of expression of CTGF. The third possible mechanism may be that TMP can improve the expression of MMP13mRNA to promote degration of collagen. |
