OBJECTIVESAt present, Staphylococcus epidermidis is one of the ahead four bacterias causing nosocomial infections, and biofilm is the major causative agent. To treat these infections, a large amount of antibiotics is used, which easily causes the bacterias to have drug resistance. Chinese material medica, unlike the antibiotics, is a predominance of new drug exploiture in our country. This experiment plans to select the sensitive monomer of Chinese material medica, explore the mechanism of influence on Staphylococcus epidermidis biofilm genes through monomer of Chinese material medica, then use this way in clinic detection to provide new method in clinical treatment.METHODS1. Using the Staphylococcus epidermidis ATCC35984 to construct biofilm in vitro, and selecting the better monomer of Chinese material medica .2. Using the model of Staphylococcus epidermidis ATCC35984 biofilm, to explore how monomer of Chinese material medica influencing key gene (icaA gene and agr gene) of Staphylococcus epidermidis by the way of fluorescent quantitation PCR, comparing Norvancomycin, Ciprofloxacin, Erythromycin.3. Using Staphylococus epidermidis from clinic to construct biofilm in vitro ,and to explore the influence on icaA gene ande agr gene by monomer of Chinese material medica through fluorescent quantitation PCR, comparing Norvancomycin, Ciprofloxacin, Erythromycin also. So that we can master the application of monomer of Chinese material medica in clinic.RESULTS1. We selects monomer of Chinese material medica——berberine hydrochloride from Chinese gall,Coptis chinensis,Baical skullcap root,Phellodendron amurense,which is the key component in Baical skullcap root and have better effectiveness to Staphylococcus epidermidis bioflm.2. The effectiveness of berberine hydrochloride influencing Staphylococcus epidermidis icaA gene and agr gene is less better than antibiotics. And Using different chroma of berberine hydrochloride and Norvancomycin, Ciprofloxacin, Erythromycin to the biofilm, The influences on icaA gene and agr gene are attenuated with the increasing chroma of berberine hydrochloride.3. The eight clinical bacterias, which can form biofilm , are 100% sensitiveness to Norvancomycin,100% drug resistance to Erythromycin, and 76% drug resistance to Ciprofloxacin. That is to say, taking out the effectiveness of Staphylococcus epidermidis biofilm, Berberine hydrochloride is weaker than Norvancomycin, but stronger than Erythromycin and Ciprofloxacin , especially Ciprofloxacin.CONCLUSIONS1. Berberine hydrochloride, as a common anti-bacteria Chinese material medica, is sensitive to Staphylococcus epidermidis biofilm.2. The influence on key gene of Staphylococcus epidermidis by Berberine hydrochloride is weaker than antibiotics (Norvancomycin, Ciprofloxacin, Erythromycin), and can not be combined with these antibiotics (Norvancomycin, Ciprofloxacin, Erythromycin), because the effectiveness is antagonism.3. when treating the infection caused by Staphylococcus epidermidis biofilm, the antibiotic can be used firstly. But if this bacteria is strongly resistance to antibiotic, the berberine hydrochloride should be considered.
