Objectives: To investigate the effects of salidroside on proliferatioa,activation,expression of ROCK-ⅠmRNA and TIMP-1mRNA in HSC-T6 cells, and to elucidate the molecular salidroside against hepatic fibrosis with Y-27632 as the positive control .Methods: HSC-T6 cell line was employed as the model for following study. MTT colorimetric assay was used to detect proliferation of HSC-T6 cell. The effects of salidroside and Y-27632 onα-smooth muscle actin(α-SMA)expression was analyzed by immunocytochemical staining.The effects of salidroside on ROCK-Ⅰand TIMP-1 mRNA expression in HSC-T6 cell was detected by ISH.Results:(1).Salidroside(5,10,20μmol/L)and Y-27632 (5,10,20μmol/L) had a dose-dependent increased inhibition effect on TIMP-1mRNA respectively(P<0.05).(2).The concentration of 20μmol/L salidroside and 20μmol/L Y-27632 could downregulateα-SMA protein expression of HSC-T6 cell, and the activation of HSC-T6 cell was inhibited.(3).Salidroside(20μmol/L) and Y-27632(20μmol /L) had a inhibition effects on ROCK-ⅠmRNA in HSC-T6 cell respectively(P<0.05)。Conclusions:Salidroside can inhibit TIMP-1mRNA,ROCK-ⅠmRNA expression in HSC-T6 cell, the mechanisms of which may be salidroside′s inhibition effect on Rho/ROCK signal transduction pathway。...
