Objective:To investigate the effects of curcumin on expression of MMP-2 and on preventing left ventricular remodeling in postinfarction rats.Method:100 male S-D rats were randomly divided into sham- operation group(n=10) and MI group(n=90). MI model was established by ligation of left anterior descending branch of coronary artery,rats surviving 24 hours were randomly divided into MI group (MI), high dose curcumin treatment group(MI-CH),middle dose curcumin treatment group(MI-CM), low dose curcumin treatment group(MI-CL) and solvent treatment group (So-MI). High, Middle and low dose curcumin treatment group were administered respectively curcumin 200,100, 50mg/kg.d , solvent treatment group were administered with liquor of 6 percent alcohol and Polyglycol 400 2.5ml/kg.d by intraperitoneal injection.Haemodynamics parameter, left and right ventricular weight index, collagen volume fraction and expression of myocardium MMP-2 examined by immunohistochemical analysis were measured at the end of the forth week.Result:①Compared with sham-operation rats,LVMI,RVMI,CVF, LVEDP and expression of MMP-2 were significantly increased (P<0.01), while LVSP,±dp/dt were significantly decreased(P<0.01) at the end of the forth week after AMI;②Myocardial infarct size of the AMI rats has no difference;③After curcumin treatment for four weeks, LVMI, RVMI,CVF and expression of MMP-2 were significantly decreased (P<0.01)and showed a dose-dependent manner. Whereas LVSP and±dp/dtmax were significantly increased (P<0.05).LVEDP was decreased by high and middle dose of curcumin treatment(P<0.01).Conclusion:①LV remodeling was occurred after AMI which manifested as thinness and fibrosis in infarct zone of LV, partly as the formation of ventricular aneurysm;collagen deposite and interstitial fibrosis in non-infarct zone and deteriorated Haemodynamics was followed.②Expression of MMP-2 in postinfarction rats were increased, which indicated that MMP-2 participates in LV remodeling after AMI;③curcumin effectively and dose-dependently prevent left ventricular remodeling and ameliorate function of LV by decreasing collagen deposite and expression of MMP-2.
