Objective:To investigate the relationship between the C491T and C523T allele polymorphism ofβ2-adrenergic receptor (β2-AR) gene and the essential hypertension ( EH ) in mengolian of Xinjiang.Combining with the phenotypic data, the possible suspectible marker of EH in mengolian was determined.Methods:Using randomiazed sampling methods of cross-sectional study, a population-based case-control study was carried out. The population was selected randomly from the pasturing area aged 30 to 70 years in Xinjiang province. Genomic DNA was extracted from the peripheral blood leukocytes by using Bio Basic Inc-BS474 Genome DNA Kit. Polymerase chain reaction and restriction fragment length polymorphism methods (PCR-RFLP) were used to detect the C491T and C523T polymorphism ofβ2-AR gene in mengolian including patients with hypertension and normotensive controls. Through observing the three genotypes distribution of C491T and C523T polymorphism between hypertensive group and normotensive group, the possible relationship between C491T and C523T polymorphism and hypertension of mengolian was analyzed.Results:(1)On C491T polymorphism ofβ2-AR receptor gene in the population, the frequencies of the genotype CC,CT were 98.37% and 1.63% respectively,no TT was found .The genotypic and allelic frequencies were in Hardy-Weinberg equilibrium (χ2=0.089, P=0.995). These two genotypic frequencies were 99.18% and 2.08% respectively in essential hypertensives, while no CT in normotensives. The allelic frequencies of C,T were 99.18%,0.82% and 100%,0% in two groups and there is no significant differences between these two groups(P﹥0.05)。In addition, systolic blood pressure and diastolic blood pressure had no significant difference in the groups classified according to genotypes(P﹥0.05).(2)On C523T polymorphism ofβ2-AR receptor gene in the population, the frequencies of the genotype CC ,CT and TT were 31.56% , 55.05% and 13.39% respectively . These three genotypic frequencies were 26.37%,65.17% and 8.46% respectively in essential hypertensives. The allelic frequencies of C,T were 59.02%,40.98% and 58.96%,41.04% in two groups and there is no significant differences between these two groups(P﹥0.05)。In addition, systolic blood pressure and diastolic blood pressure had no significant difference in the groups classified according to genotypes(P﹥0.05).(3) The female mengolian carrying CC genotype of C523T polymorphism have siginifidantly higher SBP and DBP reading than those who carrying TT genotype(P value were 0.01,0.025 respectively). (4) The distribution of three genotypes composed of C523T polymorphism was different between hypertensive group and the control group no statistical differences was found in male mengolian( P value were 0.334,0.394 respectively)Conclusion: (1)The C491T polymorphism ofβ2-AR gene was not related with essential hypertension ,which implicates it may not play an major role in the pathogenesis of hypertension in mengolian of Xinjiang.(2) The C523 allele ofβ2-AR receptor gene may be one of a genetic suspectibility factors for the hypertension of mengolian in Xinjiang,but T523T genotype might be a protective fator for the hypertension of mengolian. Mengolian carrying C523C genotype, especially for female mengolian are apt to develop essential hypertension.
