Objective:To explore the chinese medical disease mechanism of Ulcerative Colitis (UC) and put forward a new academic viewpoint.Through the research and experiment,we could observe QiXianAnChangFang GXACF's effects on UC , the effect of NF-κB and demonstrate its functional mechanism by comparing which with SASP's effects.Methods:The animal experiment copied a rat UC model induced by rinitrobenzene-sulfonic acid(TNBS) and alcohol. 48 health apinoid male SD rats,mean weight of 200~220g,were divided randomly into 6 groups(8 of one group).Selected 1 group as blank contrast group.Other 5 groups were made models by TNBS/alcohol .Then filled the normal group rats with normal saline by intragastric administration for 14 days;and named other 5 groups as:model contrast group,high-dose group, middle-dose group,low-dose group and SASP group,filled respectively with normal saline,High-dose group decoction, middle-dose group,low-dose group and SASP group by intragastric administration for 14 days.Observed the change of the general condition .The day after the model being made,we cured them with GXACF and SASP respectively,observing their effects by setting up comparative model group. The experiment contained three components.ExperimentⅠwas about GXACF's effect on generla status(spirit state,activities,hair,luster,eating and living status) and their pathological changes during the period;ExperimentⅡwas about GXACF's effect on IL-2 IL-4 activity and TNF-αcontent in serum;ExperimentⅢwas about GXACF's effect on NF-κB in tissue.Through these experiments we could explore UC disease mechanism further.Results:1 ExpermentⅠ:Comared with normal group,the other group's general status changed differently.The spirit state,activities, hair luster and foodintake of high-dose(330.00±21.38)and middle-dose(347.50±16.69) group rats were better than other groups and their weight also increased rapidly (P<0.05).SASP group(295.00±9.26)and low-dose group(298.75±13.56)were worse and model group (270.00±35.05)was the worst (P<0.05).CMDI:All of curing groups and SASP group were better model group(2.673±0.615).The high-dose(0.566±0.198)and middle-dose group(0.684±0.225) were obviously better than SASP(1.498±0.387)and low-dose group(1.602±0.152)(P<0.05).the difference of rats was statistical significance.The difference was no statistical significance between high-dose and middle-dose group(P>0.05).The low-dose group and SASP group had no statistical difference yet(P>0.05).The method of copying model induced by TNBS and alcohol was simple and feasible and its achievement ratio was high.the model's pathology changes were similar to human's.Model group rats'colonic mucosas were unabroken,with congestion, edema and infiltrating of inflammatory cells.SASP group and three Chinese herbal groups'colonics mucosas were unabroken,without such appearance above-mentioned.2 ExperimentⅡ:The contents of serum TNF-α(57.75±20.44) and activities of serum IL-2 (48.98±14.91) in high-dose group and The contents of serum TNF-α(63.36±17.85)and activities of serum IL-2 (51.02±10.93) in middle-dose group were lower than low-dose group serum IL-2 (65.73±5.21) TNF-α(94.53±31.83),SASP group serum IL-2 (64.28±6.82) TNF-α(92.35±31.55)and model group serum IL-2 (78.38±8.23)TNF-α(124.88±41.22),activities of serum IL-4 in high-dose group(30.10±5.16) and middle-dose group(26.20±7.89) were higher than low-dose group(19.37±6.49),SASP group(19.49±5.01)and model group(12.73±6.04) (P<0.05).they had no statistical difference compared with normal group(P>0.05).The difference was no statistical significance between high-dose and middle-dose group (P>0.05).3 ExperimentⅢ:The level of NF-κBp65 in modle-dose group(4.875±1.246)were higher than the normal group(1.375±0.518)(P<0.01).The level of NF-κBp65 in GXACF and SASP group(2.625±1.061)were lower than the modle group(4.875±1.246)(P<0.01).The difference was significance between low-dose(2.500±0.926)and normal group(P>0.05).Conclusion:The study pointed out that Pi weakness,Shi gravity and stagnant blood were UC's mechanisms.We made up a therapy principle of improving Pi,dispeling Shi and disappwaring carbuncle and GXACF.The research showed that GXACF could improve UC rat's nurture condition and promote their intestines'digestion.Meanwhile it could improve rat's weight apparently.Furthermore,it could dicrease the level of IL-2 and TNF-αof UC rats,promoting IL-4 activity,dicrease the level of NF-κB,reducing cell inflammation,renovating intestinal mucosa,curing of canker.The study proced that GXACF had great protective effect of UC rats.UC mechanism maybe relates to some inflammatory factor.
