Object To observe the expression of EGFR and CTGF in intrahepatic bile duct epithelial cells of infantile biliary tract obstruction and to analyze the relationship of them with hepatic fibrosis of infantile biliary tract obstruction.Methods 25 hepatic samples as the testing group were bioptically collected from the infantile biliary tract obstruction patients in the Second Hospital of Hebei Medical University. 5 normal hepatic samples of control group were collected from the less than 180 days old autopsy patients. All samples were fixed with 10% formalin, embedded with paraffin, and continuously sliced into 5μm sections. After stained by Masson method and observed under microscope, the rabbit anti-EGFR monoclonal antibody and rabbit anti-CTGF multiclone antibody were selected and the immunohistochemical experiment was finished to all samples.Results The expressions of EGFR and CTGF in testing groups are obviously evident compared with control group. In addition, in the testing groups, the positive distributions of EGFR and CTGF expression in biliary atresia group are more eminent than that in cholestatic syndrome group. ( statistical data…..) The pathology study indicates that, in the testing groups, the bile canaliculus hyperplasia and hepatic fibrosis are obvious compared with control group. But in the testing groups, the degrees of bile canaliculus hyperplasia and hepatic fibrosis are different (statistical data…..). In testing groups, the expressions of EGFR and CTGF are positively correlated with the degree of bile canaliculus hyperplasia (statistical data…..). The degree of bile canaliculus hyperplasia is positively correlated with the degree of hepatic fibrosis (….). And lastly, the expression of EGFR is also positively correlated with that of CTGF (……).Conclusion EGFR is a membrane receptor which plays a very important role in the hepatic fibrosis of infantile biliary tract obstruction. And CTGF as an important cytokine also takes part in this course. This study shows that EGFR and CTGF are expressed obviously in intrahepatic bile duct epithelial cells of infantile biliary tract obstruction. The degrees of their expression are positively correlated with the degrees of bile canaliculus hyperplasia and hepatic fibrosis. And the expression of EGFR is also positively correlated with that of CTGF. These data indicates that EGFR and CTGF might be the crucial factors in the hepatic fibrosis of infantile biliary tract obstruction, and might be some of reasons for the special rapid hepatic fibrosis progress of infantile biliary tract obstruction.
