Study On Relationship Between Types Of Differentation Of Symptoms And Signs Of Traditional Chinese Medicine And Laboratory Data In Systemic Lupus Erythematosus

Objective:To investigate the relationship between objective-parameters and Traditional Chinese Medical(TCM)syndrome types in systemic lupus erythematosus (SLE),to make up some objective parameters for TCM syndrome types in SLE,and further to elucidate pathogenesy of molecular machanism in SLE.Methods:82 cases with SLE were divided into four groups according to TCM syndrome types:Qi Xue Re Sheng Xing(QXRSX),Pi Shen Yang Xu Xing (PSYXX),Qi Yin Liang Xu Xue Yu Xing(QYLXXYX),Qi Zhi Xue Yu Gan Yu Xing(QZXYGYX);healthy control group had 31 healthy people.Then to draw peripheral blood and to do next experiments.1 The levels of mRNA expression of IL-10,IL-18 and Fas in eripheral blood monouclear cells(PBMC)were detected by SYBR Greenâ… real-time RT-PCR,and analyzed distribution regularity of them in four TCM syndrome types in SLE.2 The levels of IFN-γ,IL-2,IL-4,IL-10 and IL-18 in serum were detected by ELISA, and analyzed their distribution pattern in four TCM syndrome types in SLE. parameters3 To investigate the relationship between the clinical routine tests such as WBC,PLT, routine test ESR,complements(C₃,C₄),IgG and autoantibody(ds-DNA,S_m,RNP,SSA,SSB)and TMC syndrome types in SLE.4 To investigate the relationship between the levels of mRNA expression of IL-10,IL-18 and Fas in PBMC and clinical routine tests,the relationship the levels of IFN-γ, IL-2,IL-4,IL-10 and IL-18 in serum and clinical routine tests in SLE.Results1 The levels of mRNA expression of IL-10,IL-18 and Fas in all patients with SLE were significantly higher than that of heathy control group(p＜0.01 or p＜0.05).The level of IL-10 mRNA,IL-18 mRNA and Fas mRNA decreased by turns from types of QXRSX,PSYXX,QYLXXYX,to QZXYGYX.The three factors had one common feature in TCM syndrome types:types of QXRSX and PSYXX were significantly higer than types of QYLXXYX and QZXYGYX,and comparison of them had statistical significance(p＜0.01);However comparison of QXRSX and PSYXX, QYLXXYX and QZXYGYX had no statistical significance(P＞0.05).2 The levels of IFN-γ,IL-4,IL-10 and IL-18 in serum of the patient with SLE were significantly higher than that in control group(p＜0.01),but the level of serum IL-2 in SLE was more lower than that in control group(p＜0.01).The comparison in TCM syndrome types:(1)The level of IFN-γ:types of QXRSX and PSYXX were significantly higher than types of QYLXXYX and QZXYGYX(p＜0.01);However comparison of QXRSX and PSYXX,QYLXXYX and QZXYGYX had no statistical significance(P＞0.05).(2)The level of IL-2:types of QXRSX and PSYXX more lower than types of QYLXXYX and QZXYGYX(p＜0.01);Comparison of QXRSX and PSYXX had no statistical significance(P＞0.05);Comparison of QZXYGYX with QXRSX,PSYXX and QYLXXYX had significantly statistical significance(P＜0.01 or P＜0.05).(3)The level of IL-4:types of QXRSX and PSYXX were significantly higher than types of QYLXXYX and QZXYGYX(p＜0.01); Comparison of QXRSX and PSYXX had no statistical significance(P＞0.05); Comparison of QZXYGYX with QXRSX,PSYXX and QYLXXYX had significantly statistical significance(P＜0.01)).(4)The level of IL-10:types of QXRSX and PSYXX were significantly higher than types of QYLXXYX and QZXYGYX(p＜0.01);Comparison of QXRSX and PSYXX had no statistical significance(P＞0.05);Comparison of QZXYGYX with QXRSX,PSYXX and QYLXXYX had significantly statistical significance(P＜0.01 or P＜0.05).(5)The level of IL-18:types of QXRSX and PSYXX were significantly higher than types of QYLXXYX and QZXYGYX(p＜0.01);However comparison of QXRSX and PSYXX,QYLXXYX and QZXYGYX had no statistical significance(P＞0.05).3 With regard to counts of WBC peripheral blood of patients with SLE,types of QXRSX significantly lower than those of in types of PSYXX,QYLXXYX and QZXYGYX(p＜0.05));And ESR of QXRSX and PSYXX higher than that of QYLXXYX and QZXYGYX(p＜0.05));Comparison of counts of PLT in four types had no statistical significance(P＞0.05).The level of C₃ in QXRSX with SLE lower than that in PSYXX,QYLXXYX and QZXYGYX.For the level of C₄,types of QXRSX more lower than QYLXXYX and QZXYGYX(p＜0.05)),but comparison of QXRSX and PSYXX had no statistical significance(p＞0.05).The concentration of IgG in blood serum IgG in types of QXRSX more higher than that in types of PSYXX,QYLXXYX and QZXYGYX.Comparison of positive ratio of autoantibody (ds-DNA,S_m,RNP,SSA,SSB)had no statistical significance in four syndrome types(P＞0.05).4 By studying the relationship between anti-ds-DNA antibody and the mRNA expression of IL-10,IL-18 and Fas in PBMC of patients with SLE,find the level of mRNA expression of IL-10,IL-18 and Fas in patients with positive anti-ds-DNA antibody more higher than those of in patients with negative anti-ds-DNA antibody. As well,by studying the relationship between anti-ds-DNA antibody and the level of IFN-γ,IL-2,IL-4,IL-10,IL-18 in blood serum in SLE,find the level of IFN-γ, IL-4,IL-10,IL-18 in patients with positive anti-ds-DNA antibody more higher than those in patients with negative anti-ds-DNA antibody,however result of IL-2 was contrary.
