Objective:The project is funded by Tianjin Natural Science Foundation,which includes two parts of experiment and clinical.Part of experiment aims at establishing the method of PROD to detect the level of serum 1,5-AG which by Tianjin blood center. This study responses for clinical part,which aims at detecting the level of serum 1,5-AG,HbAlc,FMN in three groups,which divided into NGT,IGT,T2DM by the WHO standard of diagnosis,and clarify the significances of 1,5 anhydro-D-ghlcitol in early diagnosis of diabetesMethods:1.In total of 360 case including:a number of 240 patients(mean age54.18±4.48 years,males 165,female 75)who were selected consecutively from May 2006 to May 2007 in endocrine clinic of TianJin Medical University General Hospital,an oral glucose toleranced test(OGTT)was performed;and a number of 120 person(mean age50.08±3.43 years,males 70,female 50)who were examined in regular.Physical examination were given on every subject with history of disease recorded,blood sample taken.2.An oral glucose toleranced test(OGTT)was detected with the glucooxidase method.Serum insulin was detected with the radio immunity method. Acording to the results,all subjects were divided into three groups,as NGT 120,IGT 134,T2DM 106.3.Serum 1,5-AG level in three groups(NGT,IGT,T2DM)was detected with been set up measurement of 1,5-AG.Serum HbAlc was detected with the liquid low pressure gradient elution method.Serum FMN was detected with the immumal turbidity method.4.Collect date of the above three crowd,analysis the changing curve of 1,5-AG,HbAlc,FMN in NGT,IGT,T2DM.5.All date was analyzed with SPSS 11.5 software.When comparing several samples,analysis of variance(ANOVA)was used.Two samples were performed using t test. Results:1.Serum 1,5-AG level showed decline trend significantly in three groups(NGT, IGT,T2DM),respectively level is 105.56±53.46,49.42±40.07,9.59±11.64,and the concentration have statistical significance(P<0.01).2.In three groups(NGT,IGT,T2DM),serum HbAlc level respectively is5.35±0.56,5.46±0.57,7.16±0.96,the concentration between NGT and IGT has no statistical significance(P>0.1),while the concentration between NGT and T2DM, IGT and T2DMhave statistical significance both(P<0.01).3.In three groups(NGT,IGT,T2DM),serum FMN level respectively is 0.245±0.019,0.247±0.021,0.357±0.018,the concentration between NGT and IGT has no statistical significance(P>0.1),while the concentration between NGT and T2DM, IGT and T2DM have statistical significance both(P<0.01).4.Investigating the difference of serum 1,5-AG,HbAlc,FMN in IGT and T2DM,The conclusion is that serum 1,5-AG has the highest sensitivity in diagnosis of diabetesConclusion:1.In the early diabetes(IGT),Serum 1,5-AG level has showed decline trend which has statistical significance.Moreover,1,5-AG has the highest sensitivity in the diagnosis of diabetes.2.While serum HbAlc level and FMN level have variances of statistical significance in patients who have been diagnosed as diabetes(T2DM). Moreover combined with 1,5-AG,the sensitivity in the diagnosis of diabetes with HbAlc and FMN has highly improved.3.1,5-AG had important value in evaluating treatment effects in patients with diabetes,which was worth spreading for clinical application.
