The Effect Study Of Artesunate Combined With Cisplatin On Melanoma In Mice

Objective:To investigate the antitumor effect of artesunate(Art)combined with cisplatin(DDP)on melanoma.Methods:1)Growth inhibition induced by Art on melanoma B16F₀ cell line was assessed by MTT method;then the cytotoxic effect of DDP combined with small-dose Art on melanoma cell was assessed.2)Thirty-two C57BL/6J mice,subcutaneously transplanted with malignant melanoma tissue,were randomly divided into 4 groups:(1)NS,0.2ml/d;(2)Art,15mg/kg.d;(3)DDP, 2mg/kg.d,and(4)combined group,15mg/kg.d Art and 2mg/kg.d DDP.From the third day after transplantation,every mouse was administered ip once for consecutive 7 days.The longest and the shortest diameters of each tumor were measured.All mice were sacrificed a week after the end of administration. Executed subcutaneous tumors were weighed to calculate the inhibition ratio of tumor.Results:1)Artesunate suppresses the melanoma B16F₀ cell proliferation,and the inhibitory effect is dose-dependent.When the concentration of Art is between 0.39μg/ml and 0.19μg/ml,there is no significant difference between Group Art and the control group in vitro(P＞0.05).2)When the concentration of DDP is larger than 7.5μg/ml,there is no significant difference between Group DDP and the combined group in vitro (P＞0.05).But when the concentration of DDP is decreased,the cytotoxic effect of DDP combined with Art is significantly stronger than DDP,there is significant difference between Group DDP and the combined group(P＜0.01).3)There is no significant difference between the mean tumor weight in Group NS(1.97g)and Group Art(1.66g)(P＞0.05).There is significant difference between the combined group(0.75g)and Group DDP(1.12g)in C57BL/6J mice(P＜0.01).Conclusion:1)Artesunate suppresses the melanoma cell proliferation,and the inhibitory effect is dose-dependent.2)There is synergistic cytotoxic effect of small-dose artesunate combined with cisplatin on melanoma cell both in vitro and in vivo.
