Studies On The Active Constituents Of Swertia Nervosa(G.Don) Wall. And Content Determination Of Two Bitter Secoiridoid Glycosides

Swertia nervosa(G.Don)Wall.,which belongs to Gentianaceae,is also called "Qing Yu Cao,Si Lin Cao "et al in the folks.It is used for acute epipephysitis,sore throat,tonsillitis,icteric hepatitis,cholecystitis, gastroenteritis bacillary dysentery and urinary tract infection et al.It is reported that Iridoid glycosides and Xanthones are the majory consitituents in Swertia nervosa(G.Don)Wall.Iridoid glycosides have effects of improving function of skin,promoting hair growth,inhibiting central nerve,relieving pain,anti-inflammatory,increasing gastric secretion and anti-ectosarc.Xanthones have activities of lowing blood lipid,antioxidation,anti-ATS,protecting liver and anti-bacteria.Our studies contain two aspects,including systemic researches on chemical components to investigate the bioactive constituents and assaying of multicomponent by HPLC to controll quality and establishing the standard of medicinal materials.Several methods have been used in the processes of separation and purification including macroporous resin chromatography column(CC), silca-gel CC,pre-LC and so on.As a result,six compounds have been isolated.On the basis of physical-chemical properties and spectral evidences,their structures were elucidated as:swertiamarin(â… ), gentiopicroside(â…¡),norswertianolin(â…¢),bellidifoloside(â…£), demethylbellidifolin(â…¤)and bellidifolin(â…¥).Among them,compoundsâ…¡,â…¢andâ…£are isolated from this plant for the first time.We also found a way to measure the contents of Swertiamain and Gentiopicrin together.Methodological evaluation shows it is a reliable approach having some advantages as convenient,efficient,sensitive and being-reproducibility.Swertiamain and Gentiopicrin are the main active components in Swertia,Hunan Standard of Traditional Chinese Medicinal Materials(2005 edit)has the TLC-identification of Swertiae mileensis by using Oleanolic acid and Swertiamain as control article excepting for assaying.The means measuring the contents of Swertiamain and Gentiopicrin together in Swertia nervosa and Swertia davidi are reported for the first time,which not only supply a reference on investigating Swertia nervosa and Swertia davidi as substitutes for the other plants in Gentianaceae,but also offer the scientific evidences for further exploitation of Iridoid glycosides and experimental basis of artificial cultivation and development of swertia in the west of Hunan.
