Objective:(1)To set up a rat animal model for donor liver havest.(2)To investigate the difference of the level of TNF-αin the graft caval effluent after flushing of the liver with the cold preservation time changed.(3)To investigate the relationship of the level of TNF-αin the graft caval effluent after flushing of the liver and the graft quality,and to seek a potential method of assessment graft quality before implantation.Method:The research target are healthy male SD rats.(1)The animal model is liver harvest of rat liver transplantation,and preserve the liver in 0-4℃UW fliud.(2)Divide the experimental animals into seven groups according to the time change,collect the samples and examined liver function and TNF-αconcentration,pathology examination was done for the donor liver.(3)All the experimental data was analysised by statistic method.Results:(1)With the same warm ischemic time,the TNF-αconcentration increased with the cold preservation time.(2)We find obvious morphological changes through the pathology examination in the donor liver.(3)The TNF-αconcentration in the graft caval effluent is correlation with the ALT and AST level..Conclusion:(1)TNF-αplays a important roles in liver ischemic repurfusion injure,and is a main cytokine to mediate injure.(2)The liver function test and pathology examination is valuable for evaluating donor quality.(3)Through this experiment we consider that the TNF-αconcentration in the graft caval effluent is a potential index in evaluating liver donor quality.
