Object:To study the polymorphism of HLA-A,B,DRBI loci in Hunan Han population and to compare the distribution of HLA-A,B,DRB1 alleles and haplotype with the other populations.To establish the HLA genotyping database of Hunan population.Methods:The HLA-A,B,DRB1 low resolution genotyping(n=3664) was done by the polymerase chain reaction with sequence specific primer technique.The allele and haplotype frequencies of HLA-AB,HLA-BDR,HLA-ABDR were obtained by a square-root method and 2×2 table.The genetic distance among the 12 populations were calculated by the Nei formula.The cluster analysis and phylogenetic tree were figured out by weighted pair-group method using arithmetic averages(WPGMA) method.Result:63 alleles were detected in total.The most frequent HLA-A,B,DRB1 alleles in Hunan population are HLA-A*11(34.42%),A*02 (31.88%),A*24(18.01%),A*33(6.18%),B*40(23.70%),B*46 (17.76%),B*15(13.99%),B*13(8.60%),DRB*09(19.69%), DRBI*15(14.01%),DRBI*12(12.97%),DRBI*04(11.96%)and DRB 1*08(9.02%)respectively.The frequency of HLA-DRB 1*09 allele is higher than other populations in China.350 HLA-AB haplotypes,408 HLA-BDR haplotypes and 1935 HLA-ABDR haplotypes were detected. Among them,36 HLA-AB haplotypes and 48 HLA-BDR haplotypes were in strongest linkage disequilibrium.The most common haplotypes were A2-B46,A11-B60,A24-B60,B46-DR9,B60-15,B60-DR4,A2-B46-DR9,A33-B58-DR3,A11-B75-DR12 respectively.Comparing with other populations,the genetic distance between Hunan Han and Jiangxi Han is the nearest(0.0046),while Shandong Han is the farthest (0.1290).Comparing with other populations,distinct differences were found among them.2 clusters were achieved in the cluster analysis.The Hunan Han population was clustered with Jiangxi Han first of all,and then clustered with Hubei Chongqing Han,Guangdong Han respectively. The five populations belong to the Southern population.At last,the Southern Han clustered with the Northen Han,which consisted of Dalian,Xi' an,Shanxi,Henan and so on.Conclusion:1.The polymorphism of HLA-A,B,DRB1 loci and haplotype are very multiplicity.It is not too difficult for the leukemic patients to find appropriate donors of haematopoietic stem cell transplantation in Hunan.2.The HLA frequency and haplotype distribution of Hunan population is quite different from Causian,Blackman,Korean,Northen Hart and other populations.It is similar to Han population in South China,but it has its own characteristics.3.There are 2 clusters achieved in the analysis.Hunan Hart belong to Southern Han population.Hunan Han and the other Hans constitute Chinese Han together.4.The HLA genotyping database of Hunan population has been established and the study of HLA can provide the invaluable tools and backgroud for the anthropological and historic study of Hunan area.It also can be considered as the normal refrence value of Hunan population for the study of the association between HLA and disease.
