Objective To study the effect of reversal multidrug resistance on human lung adenocarcinoma cell line A549/CDDP by human recombinant interleukin-2 andα-interferon.Methods Human adenocarcinoma of lung cell line A549/CDDP was cultured in vitro, and was treated by different density of IL-2 andα-IFN. drug sensitivity of IL-2 andα-IFN was analyzed using the MTT assay, intracellular accumulation rhodamine by P-glycoprotein function expression was examined by fluorospectrophotometry.Results1. The intracellular accumulation of rhodamine is difference significant in A549/CDDP cell line between control group (P<0.05) after combination treatment by IL-2 andα-IFN.2. The in tracellular mean rhodamine of A549/CDDP cell line were treated by IL-2 andα-IFN after 48h, their combination treatment group is: IL-2 group is 4.82;α-IFN group is 4.25; EL-2 combination withα-IFN group is 9.13; control group is 2.38. There is significant difference (P<0.05).3. The intracellular mean fluorescence of rhodamine in A549/CDDP cell line of is 5.98,8.90,3.03 after IL-2 andα-IFN, and control group is 15.23(P<0.05).Conclusion IL-2 andα-IFN can reverse the MDR. Its possisblemechanism is inhibiting function of P-gp and increasing the accumulationof intracellular chemotherapeutic CDDP in A549/CDDP cells.
