Objectives:This experiment is to investigate a proteome profile of normal CSF of Chinese children by two-dimensional polyacrydamide gel electrophoresis(2-DE),and to sieve the disease- specific proteins(DSPs) to provide clues for the early diagnosis and further therapy of Staphylococcus epidermidis meningitis(SeM).Methods:2 SeM patients and 2 normal control patients were included in this study.4 ml CSF of each patient was obtained.The whole proteins of CSF were got through trichloracetic acid(TCA),then were separated with immobile pH gradient(IPG)2-DE technology.The stained 2-DE gels were scanned on the Imagescanner and analyzed with PDQuest software.Then the different proteins spots were detected between the SeM and normal children.5 clearly discerned spots were chosen to carry out Matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF-MS)analysis to obtain 5 Peptide mass fingerprintings(PMF).Finally,we get the basic information of the proteins through bioinformatics to definite specific proteins.Results:Mean protein spots of the 2-DE gels were 438 and 425 with SeM and normal children respectively.62 different protein spots were screened between the SeM and normal group,of which 6 protein spots showed up-regulation and 19 spots down-regulation for 2 folds,25 sopts only occurred in normal CSF and 12 spots only occurred in SeM group. MALDI-TOF-MS was used to analyze 5 differential protein spots and 5 PMF were obtained.After searching the data base,5 proteins were matched.Finally high affinity glutamate transporter and Alpha-2-HS-glycoprotein were specific expressed in SeM CSF,HP protein was expressed up-regulatedly in SeM CSF,apolipoprotein E and kallikrein 6 were expressed down-regulatedly in SeM CSF.Maybe these were disease specific proteins of SeM.Conclusions:A 2-DE profile of CSF proteome was successfully established in SeM and normal children through proteomic technique.By the differentiated analysis of these CSF 2-DE gels,the differences of CSF proteome profiles,including numbers and densities,were discovered between SeM and normal children.The high affinity glutamate transporter,Alpha-2-HS-glycoprotein,HP protein,apolipoprotein E and kallikrein 6 maybe related to the occurance and development of SeM. Meanwhile,to eatablish foundation for the early diagnosis and new medicine.
