Objective:To study the regulation of GLAST by BDNF in the retinal Müller cells of mice.Methods:(1)retinal Müller cells of mouse at postnatal 3 to 7 days were cultured by tissue inoculation.(2)Cultured retinal Müller cells were divided into two groups:①BDNF group:Cultured retinal Müller cells were maintained in different concentrations of BDNF(50ng/ml,75 ng/ml,100 ng/ml,125 ng/ml and 150 ng/ml)for 24 h in vitro;②Control group:Cultured retinal Mǜller cells without BDNF.(3)The expression of GLASTmRNA in the retinal Müller cells was semi-quantified by RT-PCR.(4)The expression of GLAST protein in the retinal Müller cells was detected with immunocytochemical staining.Results:(1)The expressions of GLAST mRNA was up-regulated as the increase of the concentrations of BDNF.The 100 ng/ml BDNF caused a maximal increase in GLASTmRNA expression(grey scalue:1.96±0.13).(2)Immunocyto-chemistry indicated that GLAST protein expression located in the cytoplasm and membrane of retinal Müller cells.The effect of 100 ng/ml BDNF on the expression of GLAST protein were higher than that in the control group,which were significant differences among these five groups(P<0.05)Conclusion:BDNF can increase GLAST activity through up-regulating the expression of GLASTmRNA and protein.
